Forsythia is the fruit of Forsythia, a member of Oleaceae plants.Most of the Forsythia plants grow in mountains, and they have been cultivated in many places,such as Shanxi, Henan and Shanxi. Forsythia is bitter,slightly cold, and it is one of the commonly used detoxification drug. Forsythia has the disinfection and detumescence effects, it is used for such symptoms:ulcer, mastitis, scrofula, anemopyretic cold, erysipelas, epidemic febrile disease, hige fever polydipsia, anuria and coma eruption. Along with depth research on Forsythia in recent years, it is found that forsythiaside A-J play a major roles in Forsythia’s pharmacological effects, and forsythiaside A has the highest content among them.Firstly, this study prepares forsythoside A as reference. Using70%ethanol solution extracts Forsythia suspense, then integrated using normal phase silica gel column chromatography, polyamide column chromatography and RP-C18silica gel column chromatography to separate and purify70%ethanol extract, then obtain a compound. It’s purity is above98%Analyzed by HPLC and determined as forsythoside A by13C NMR.Secondly, this study designed a novel total Forsythia glycosides extraction and purification craft:first of all, Using70%ethanol to percolated extract fruits of Forsythia suspense; then using petroleum ether: ethyl acetate(1:1) to extract concentrated liquid to except little polar impurities; finally, adding15%ammonium sulfate to the liquor and extracting with ethyl acetate:methanol(95:5), the evaporated extract is total Forsythia glycosides.
