Deerskin is one of the most important components of the deer. Because thedeerskin has many characteristics, such as soft, tough, wear-resisting, is widelyapplied in leather goods and it is a kind of very high-end leather material.And becausehistorical records deerskin is traditional Chinese medicine which can treat kidneyempty,so the deer was applied to the development of deer skin glue and many healthynutritional products.But the reasearch of the extraction of deerskin collagen and itsprouction is few. In this paper, we use the method of ultrasonic assisted the deerskincollagen extraction and the optimal process is determined. In this paper, the collagenof enzymatic hydrolysis is raw separated and is developed into a new kind of redjujube and collagen juice. Specific research results are as follows:1. Ultrasonic assisted extract the collagen and its text conditions optimizationThrough the experiment we select the method of ultrasonic to extract the deerskincollagen, and select the neutral protease enzymolysis the deerskin.The proteinextraction rate is as the standard to the ultrasonic deerskin collagen enzyme solutionof the optimal material liquid ratio is1:10, optimum temperature is50℃, the mostsuitable time is40min, the optimum power is200W.Parameters were optimized by response surface combination design, regressionequation is:Y=54.20+2.74X1+8.23X2+2.91X3+1.35X4+2.22X1X2-1.27X1X3+1.88X1X4+1.08X2X3-1.45X2X4-2.48X3X4-2.18X12-11.59X22-4.75X32-3.21X42Best combination is: the extraction yield of collagen is the largest as high as57.22%when the material liquid ratio is1:14.8, ultrasonic temperature is54.3℃,ultrasonic time is42.8min,and the ultrasonic power is208.5W2.the sepration of collagen and its antioxidant activityBy gel electrophoresis map shows the collagen enzyme solution, and afterultrasonic assisted collagen enzyme solution effect is very good, appeared a lot ofdifferent molecular weight of proteins or peptides.The enzyme collagen is separated by ultrafiltration and for each separatecomponent of antioxidant experiment research which shows hat when the molecularweight of less than20000Dalton collagen peptide has the strongest antioxidantactivity.The collagen polypeptide which has the strongest antioxidant activity has beenseparated by gel G-75, the separation results showed a two molecular weight protein.Each kind of molecular weight of the protein was studied the antioxidant test, thestudy shows that the lower molecular weight fractions has the stronger antioxidantactivity.3. The development of the red jujube and collagen liquid compound beverageThe best Volume ratio of jujube and collagen、add the best level of cane sugar,citric acid and the best amount of stabilizer. Through the experiment the parameterswere optimized by orthogonal test which shows the result: The ratio of red jujube andcollagen liquid is2:1,Sucrose content was10%, Citric acid content was0.08%. The colour of red jujube and collagen liquid compound beverage is uniform and tastedelicious,it is full compliance with national standards.
