Over the past30years since its invention in1980s, atomic force microscopy has become one of the most versatile tools in nano-scale surface science. With the development of the science and technology, it has been successfully used in many fields such as medicine, pharmacy and biological detection.Compared with optical microscopes (OMs), scanning electron microscopes (SEMs) and other surface roughness measurement instruments, atomic force microscopes have more potential for the measurement and characterization of three-dimensional (3D) micro and nano structure surfaces. Today, AFM has become an important part of the biological microscope technology, it can realize high resolution imaging and manipulation of living cells in aqua, and provide a new method for the cell biology research.In this report, the mechanism of the force between the AFM probe and the sample in the gas and liquid conditions is analyzed. The AFM principle and modes of operation are described in detail. The atomic force microscopy system is designed and set up. A large area AFM system is developed, and the scanning range can be extended from100μm to25mm while keeping a high resolution in both lateral and vertical directions.
