Shanghai poultry breeding company is the national grade base of layinghens’breeding, which mainly takes part in the work of selection, producing andpopularization in the commercial strains of laying hens which contain the Xinyanghyline layer chicken. However, although as a new good breed our countryfosters,xinyang hyline laying hens have the characters itself and the advantages inhigh egg laying performance, adaptability and strong premonition,while it’s stillnecessary strengthening the choosing and breeding works,especially in the field ofreproduction work, with the purpose of increasing producing efficiency,economiceffectiveness and decreasing the dependence on the foreign layer chicken breeds atmeanwhile.Marker-assisted selection (MAS) is one kind of new and developing breedingmethod. At the same time,the molecular genetics basic research of breedingperformance about chicken lays a foundation in the MAS’s developing more orless.Among these research, Prolactin gene, Occytevitellogenesis Receptorgene,Gonadotropin-releasing Hormone Receptor gene and Ovoinhibitor gene allwere reported being directly bound up with breeding performance,while the concreteeffect also lies on the genetic background of poultry population.In order to carry out the MAS work in hyline layer chicken of Shanghai poultrybreeding company, we mainly do the following work, and obtain series of keyachievements:(1) We use the means of PCR-RFLP to detect the genetic diversities of fourcandidate genes including the four PRL、OVR、OIH and GnRHR genes, Theconsequence makes clear that that in Xinyang hyline layer chicken,the24base pair delete/insert in the proximal promoter region-378~-355base pair of Prolactin gene,the3967base pair T/G mutation in the second introns of Occytevitellogenesisreceptor gene, the location of537base pair in the first introns ofGonadotropin-releasing Hormone Receptor gene does’t exsist heterozygote and the15328base pair C/T mutation in the fourteen extrons of Ovoinhibitor gene.(2) We use the polymorphisms of four genes above to do association study withthe eggs number of51~64week old and average egg weight of76week old, all thefour candidate genes don’t reach the significant level (P>0.05), then we choose thethree PRL, OVR, GnRHR genes to merge the genotypes and made associationanalysis with the two egg triats, while it is also not significant.This result illustrates that gene’s effect rests with group specificity, that’sdifficult to get the expected effect if we carry out the MAS work blindingly.
