Localization Of Ghrelin And Its MRNA In The Ovary And Uterus Of Mouse At Different Development Stages

Ghrelin is a kind of neuropeptides and is also known as auxin. Many studies showed the presentation of Ghrelin is in brain, stomach and intestines, so Ghrelin is also known as brain-gut peptides. In order to proclaim the function of Ghrelin in animal’s reproduction and regulation, the immunohisto-chemical PV-90002-step method and in situ hybridization were used to identify the distribution and presentation of Ghrelin and its mRNA in uterus and ovary of mouse at four development stages such as puberty, pregnancy, lactation and after lactation. The result showed that (1) Ghrelin were found in ovary at four development stages, Ghrelin positive cells were mainly found in ovocyte, interstitial cells, ovary medulla and corpus luteum in ovary at the four stages. However, their distribution and intension were different. Those positive cells were stronger in the corpus luteum of pregnancy mouse and lactation mouse. Ghrelin positive cells were mainly found in the germinal epithelium of pregnancy mouse. And based on statistical analysis the average value of Ghrelin positive cells at8-day old of pregnancy and18-day old of lactation mouse was more than the puberty mouse’s (P<0.05) those average value of positive cells at8-day old of pregnancy mouse was significantly bigger than18-day old of lactation mouse’s and the mouse after lactation(P<0.05) those average value of positive cells at7-day old of lactation mouse was significantly less than18-day old of lactation mouse’s(P<0.05) those average value of positive cells at18–day old of lactation mouse was significantly bigger than the mouse after lactation(P<0.05) the gray score of puberty mouse was significantly more than pregnancy mouse and lactation mouse(P<0.05) the gray score of pregnancy mouse was significantly less than the mouse after lactation(P<0.05) the gray score of lactation mouse was significantly less than the mouse after lactation (P<0.05).(2) Ghrelin mRNA were found in ovary at the different stages above, Ghrelin mRNA positive cells were mainly found in ovocyte, interstitial cells, corpus luteum and ovary medulla in ovary of four development stages such as puberty, pregnancy, lactation and after lactation. Those positive cells were stronger in the corpus luteum of pregnancy mouse those positive cells were also stronger in the ovary medulla of mouse after lactation. And based on statistical analysis compared with the puberty mouse and the mouse after lactation the average value of Ghrelin mRNA positive cells at pregnancy mouse was great bigger (P<0.05) the average value of those positive cells at lactation mouse was significantly bigger than the puberty mouse and the mouse after lactation (P<0.05) compared with the pregnancy mouse and lactation mouse the gray score of puberty mouse was significantly bigger (P<0.05).(3) Ghrelin were found in uterus at four development stages, Ghrelin positive cells were mainly found in connective tissue chorion, lamina proprial, basal cells and mesothelium. Ghrelin positive cells were also found in simple columnar epithelium at the uterus of pregnancy and lactation mouse. And based on statistical analysis the average number of Ghrelin positive cells at pregnancy mouse was significantly more than the puberty mouse, the lactation mouse and the mouse after lactation (P<0.05) the average number of those positive cells at the mouse after lactation was significantly less than pregnancy mouse and the lactation mouse (P<0.05) the gray score of pregnancy mouse was significantly less than the puberty mouse, lactation mouse and the mouse after lactation (P<0.05).(4) Ghrelin mRNA were found in uterus at those different stages above, Ghrelin mRNA’s positive cells were mainly found in connective tissue chorion, lamina proprial, basal cells and mesothelium. Ghrelin mRNA’s positive cells were stronger in lamina proprial. And based on statistical analysis the average number of Ghrelin mRNA positive cells at puberty mouse was significantly more than the pregnancy mouse, the lactation mouse and the mouse after lactation (P<0.05) the gray score of puberty mouse was significantly bigger than the pregnant mouse and lactation mouse (P<0.05) the gray score of pregnant mouse was significant less than the lactation mouse and the mouse after lactation (P<0.05). It was concluded that Ghrelin and its mRNA were found in ovary and uterus at four development stages and the positive function were stronger at pregnant mouse, those results showed Ghrelin may play important roles in different development stages especially the pregnancy mouse.
