After orally taking Enrofloxacin, Enrofloxacin in swine body fitted to one compartment open model, here are the main pharmacokinetic parameters:Tmax (1.86±0.73) h, Cmax (1.79±0.52)μg·mL-1, T1/2ke (4.29±1.38) h, AUC (15.26±5.73) mg·h·mL-1. By comparing the medicine generation parameters of the two drugs it shows that compared with Enrofloxacin, in swine body Enrofloxacin powder is slower absorpted and has longer Tmax and elimination half life, the area under the concentration-time curve of the two drugs are nearly the same. The relative bioavailability of orally taking Enrofloxacin powder is96.13%. It has remarkable curative effect to swine Infectious Pleuropneumonia by orally taking Enrofloxacin powder5~10mg·kg-1each time twice a day, continuously taking for5days.This study also evaluates the safety of Enrofloxacin powder being orally taken by swine the targeted animals based on VICH veterinary drug target animal security research guiding principle and the ministry of agriculture Veterinary Chemicals Safety Pharmacology Experiment Guiding Principle.24healthy pigs are randomly grouped in to4groups, they are a control group and three medicated groups orally taking drug twice a day for continuously for15days with the doses50mg·kg-1(5times the recommended dose),30mg·kg-1(3times the recommended dose),10mg·kg-1(the recommended dose) relatively. The adverse drug reactions are not observed during administration periods, the blood routine indexes such as red blood cell, platelet, hemoglobin and hematocrit have no significant differences between the periods before and after drug taking, and also does the biochemical index. The pathology of main visceral biopsy does not show any obvious pathological changes. There are no significant differences in the main viscera coefficients between control group and the different-dose-drug taking groups. This conclusion suggests that orally taking Enrofloxacin powder in the dose range of10mg·kg-1~50mg·kg-1has neither noticeable influence to swine blood physiological and biochemical parameter and organization structure nor toxic reaction. |
