Enrofloxacin is the 3rd generation Fluoroquinolones special for animals, belonging to broad spectrum bactericide. It is widely used for treatment of animals in Europe and America. But Enrofloxacin is limit used because of its natural bitterness and alkalinity for directly feeding and water drinking. To solve the Enrofloxacin’s bitterness and alkalinity problem to realize oral administration, the Microencapsulated Enrofloxacin is developed. In order to investigation the clinical effects of this type of Enrofloxacin, the in vivo pharmacokinetics of Enrofloxacin in swine is determined by HPLC, and established the swine Infectious Pleuropneumonia model by challenging bacteria seed in vitro. And then the swine diagnosised treated with different dosage of Enrofloxacin (10 mg/kg、5mg/kg、2.5 mg/kg), and injected with Enrofloxacin injection (BAYTRIL) (2.5 mg/kg) was as drug control. The results are as follows.1). Both of Microencapsulated Enrofloxacin and Enrofloxacin powder were accord with a one-compartment model with first-order absorption.2). In 10mg/kg-bw oral Microencapsulated Enrofloxacin group, the result shows Tmax was 4.55h, Cmax was 1.08μg·mL-1, AUC was 14.67 mg·h·L-1, and T1/2kβ was 4.82 h。 Compared with Enrofloxacin powder, the relative bioavailability of Microencapsulated Enrofloxacin was 96.13%. Microencapsulated Enrofloxacin showed good absorption rate and bioavailability.3). Both of 10 mg/kg and 5 mg/kg Microencapsulated Enrofloxacin effectively decreased the piglet’s death rate caused by Infectious Pleuropneumonia compared with the drug control group. And the effective rate and cure rate were up to 90%. The cure rate of 10 mg/kg,5 mg/kg group and the drug control group were significantly higher than 2.5 mg/kg group and positive control group (P<0.05); The difference of cure rate between 5 mg/kg group and drug control group was not significantly (P>0.05).4).6-48 h after medication treatment, in groups of 10 mg/kg and 5 mg/kg Microencapsulated Enrofloxacin and the drug control, significantly reduced the body temperature compared with control group (P<0.05), and significantly improvd weight gain compared with control group and 2.5 mg/kg group (P<0.01). The difference of weight gain rate between 5 mg/kg group and drug control group was not significantly (P >0.05)...
