Genetic Variability And QTL Association Mapping Of Seed Soluble Sugar Content In Chinese Landrace Population Of Soybean

The soybean is one of the major crops originated in China. The cultivated soybean was evolved from the wild soybean and soybean landraces had rich morphological variation. They are important materials for discovering new genes. The nutrients in soybean seeds contained proteins, lipids, carbohydrates, vitamins, inorganic salts and some trace elements. The soluble sugar is an important part of soybean seeds. Usually it accounts for8%~10%in soybean seeds. The soybean soluble sugar is mainly about soybean oligasacchrise which is good for our health. Study the genetic variation of seed soluble sugar content in soybean landraces. Use the SSR markers for this trait QTL association mapping and dissect their elite alleles. All of these are important for good quality breeding in soybeans.In this study,332representative soybean landraces from different eco-regions in our country were selected and one year field experiment was carried out. The anthrone colorimetric method was used to detected seed soluble sugar content in soybeans. Association mapping of the loci for this trait were done by using134SSR markers. The main results were as follows:(1) The variation of seed soluble sugar content in soybean landraces was from4.17%to12.45%. The mean of seed soluble sugar content in each ecological zones soybeans was around8%. From the measured phenotype data, we found some high quality germplasm resources. The soybeans whose Nannong number were N05758, N24151, N23598, N24019, N23662, N23642, N5456, N24307, N23644and N06534.1had higher seed soluble sugar content than11%.(2) By using the GLM model of TASSEL2.1software, We found six marks were linked to soluble sugar content QTLs when the significance level was below0.05. They were Gmes4742, Gmes4727, Satt209, Gmes6556, Gmes2824and Gmes3292. And the contribution rate of them were5.31%、8.56%、5.66%、1.37%、2.04%and1.74%respectively. And they were located on chromosome2,8,8,11,15and19(the original linkage groups D1b, A2, A2, B1, E and L) respectively. When the significance level was below0.01, we found only one marker was linked to soluble sugar content QTLs which was Gmes4727.(3) There are14elite alleles and13negative effect alleles we found which associated with seed soluble sugar content in soybeans. The more efficient alleles were Gmes4727-173、Gmes4742-266、Satt209-189、Gmes4727-182、Gmes2824-282、Satt209-192、Satt209-168and Gmes6556-258. The Nannong number of typical materials which contented these elite alleles respectively were N23598、N23642、N24019、N24307、N24151、N23548、N24268and N23662. The more negative effect alleles were Gmes3292-131、 Gmes4727-176、Gmes4742-252and Satt209-174. The Nannong number of typical materials which contented these alleles respectively were N05075.1、N05075.1、N04365and N8218.(4) By analysising the genetic structure of alleles, we found soybeans which had high seed soluble sugar content accumulated a lot of elite alleles, and soybeans which had low seed soluble sugar content accumulated a lot of negative effect alleles. The variation of seed soluble sugar content in soybean landraces of each eco-region in our country had a large range, and the composed alleles accumulated in each materials of each eco-region were different. The trend of seed soluble sugar content change in soybean landraces of six eco-regions was similar, and genetic structure of composed alleles in soybean landraces of six eco-regions were also similar. According to the composition of elite alleles in the carrier materials, we chose some excellent parents which had some kernel traits for breeding in soybeans. For example, there will be six elite alleles accumulated in the update of the cross N23598×N23642, and the phenotype data of elite alleles will reach5.114(10mg/g). There will be seven elite alleles accumulated in the update of the cross N24609×N23642, and the phenotype data of elite alleles will reach3.014(10mg/g).
