Soybean is an important food and economic crop,which is rich in plant protein,oil and carbohydrate.Its nutritional and medicinal value is more and more favored by the public.The carbohydrate content of soybean seeds generally accounts for 25%-30% of the total grain matter content,among which the soluble sugar content is as high as 16%,which has important health care function for human body.The content of soluble sugar and its components directly affect the edible quality and nutritional value of soybean.Breeding soybean varieties with different contents of soluble sugar and its components can meet the different needs of the market,and meet the special needs of some breeding.In this study,the agronomic traits,the content of soluble sugar and its components,are determined by high performance liquid chromatography(HPLC)in a recombinant inbred line(RIL)population composed of 128 lines and 6 soybean varieties with different evolution types constructed by the laboratory of soybean germplasm innovation and use of Shanxi Agricultural University.The result showed:1.The content of soluble sugar and its components showed normal distribution,which is as the characteristics of quantitative characters.The difference of raffinose between years and varieties is very significant.Correlation analysis showed that a significant positive correlation between total sugar content and sucrose content,raffinose content and stachyose content.Cluster analysis showed that all materials could be divided into four groups,the third group showed the highest sugar content in three years,and the fourth group showed high glucose content and low other components content in three years.2.The results showed that no significant difference in the contents of glucose and raffinose among wild,semi wild and cultivated soybeans,but significant differences are found in sucrose and stachyose contents.The content of sucrose in wild soybean is the lowest,and the content of stachyose in wild soybean is the highest.The analysis of the relationship between soluble sugar components and agronomic traits showed that 100 seed weight has a significant positive correlation with sucrose content,a significant negative correlation with stachyose content,and a significant negative correlation between the number of three seed pods and sucrose content.In the evolution of wild soybean,100 seed weight is an important indicator,further analysis shows that the sucrose content of soybean increased and stachyose content decreased in the process of evolution.3.The genetic models of soluble sugar components in soybean are analyzed.The results showed the genetic models of glucose in 2017,2018 and 2019 are 4MG-AI,2 MG-CE and 0 MG;the genetic models of sucrose in 2017,2018 and 2019 are 0 MG,2 MG-CE and 2 MG-CE;the genetic models of raffinose in 2017,2018 and 2019 are 2 MG-CE,2 MG-CE and 3 MG-AI;The genetic models of stachyose in 2017,2018 and 2019 are 0 MG,2 MG-CE and 4 MG-AI.The further analysis shows that sucrose and raffinose are 2 MG-CE genetic model in 2 years.4.The results of QTL mapping for soluble sugar and its components in soybean seeds are analyzed by composite interval mapping.Twenty QTLs related to soluble sugar in soybean seeds are detected in three years,which are distributed on 11 chromosomes: 1,2,3,4,6,8,10,11,12,13,17 and 20.A stable locus controlling stachyose content is detected in the region of 72.231 c M-87.740 c M on chromosome 10.A locus with high genetic variability is detected in the range of96.881 c M-97.529 c M on chromosome 11,which is related to sucrose content in 2019.5.The results of QTL mapping for agronomic traits in soybean are analyzed by composite interval mapping method.Fourteen QTLs related to agronomic traits are detected on chromosomes 2,3,6,8,11,15,17 and 18.Among them,6 loci with more than 10% interpretation rate are plant height,plant weight,grain weight per plant and 100 seed weight,which are located on chromosomes 2,11 and15.Plant height,plant weight and grain weight per plant are located on chromosome 11,and the QTLs of plant weight and grain weight per plant are similar.One pod,seed weight per plant and 100 seed weight are all located on chromosome 15.6.293 genes are annotated,among which 180 genes are polymorphic between parents.Among them,two genes related to glucose metabolism are Glyma.10g154400 and Glyma.11g136200,respectively.The work of this gene needs to be further verified.
