The purpose of this studies is to explore the protective effect of astragalus polysaccharide (APS) and Puerarin on diethylstilbestrol (DES)-induced adult mouse testis damages and to do a contrastive analysis.(DES)-induced adult mouse testis injury.80male mice were divided into normal control group,diethylstilbestrol group,3APS groups (APS low-dose group, APS middle-dose group, APS high-dose group) and3Puerarin group(Puerarin low-dose group, Puerarin middle-dose group, Puerarin high-dose group). APS low, middle, high dose group were administered40,60,100mg·kg-1of APS; Puerarin low, middle, high dose group were administered50,100,150mg·kg-1of Puerarin; The control group was given equivalent amount of normal saline. The mice in the other7groups were injected hypodermically with DES at40mg/kg BWdaily4hours after the first herbal administration, while the control was given olive oil. Administered continuously for7days and then processed. Then calculates testis weight and its organ coefficient; detectes changes in testicular pathology:detectes superoxide dismutase (SOD) activity, glutathione peroxidase (GSH-Px) activity and malondialdehyde (MDA) content of testicular; discoveres hormone levels of testosterone (T), luteinizing hormone (LH) and gonadotropin-releasing hormone (GnRH) in serum.The results showes that in the APS groups and Puerarin, compared with the DES model group, testis weight and its organ coefficient has significant differences; changes in testicular pathology has significant improvement. The antioxidant results shows that, in the each dose group of APS and Puerarin, SOD, GSH-Px activity of the testicular tissue are higher than the DES group, the difference is significant. SOD, GSH-Px activity of the Group100mg·kg-1is the highest. In the each dose group of APS and Puerarin, MDA levels of testicular tissue are lower than the DES group, the difference is significant. Among them, the60mg·kg-1group is the lowest. In the each dose group of APS and Puerarin, content of T, LH, GnRH in the serum are significantly increased. The results suggest that the APS had a palpable role in relieving the reproductive damages induced by the DES.
