Phospholipid (PL) is one of the important flavor substances in livestock and poultry meat. Foreign scholars have done many researches and conducted a series of verifications. The major compound of membrane phospholipids is phosphatidylcholine in Mammals. PC Synthesis is mainly regulated by the Kennedy way. A key enzyme in the process is CCT(CTP:phosphocholine cytidylyltransferase). Another way, synthesis of PC is catalyzed by the key enzyme PEMT (phosphatidylethanolamine N-methyltransferase). PC content in tissues of Suhuai pig was detected by HPLC. In this study, Bioinformatics analysis of PCYTIA and PEMT in pig was also focused on. PC content and mRNA relative expression level of PCYT1A and PEMT genes in different tissues of Suhuai pig were analyzed. This research revealed the synthesis of PC in tissues and established the foundation of the synthesis of PC in muscles.1Bioinformatics analysis of PCYTIA and PEMT gene and proteins.The gene sequencing, protein structure and phylogenetic relationship were analyzed by bioinformatics software. The physical and chemical properties of pig PCYTIA indicated that PCYTIA is a hydrophilic protein. There weren’t strongly signal peptide and transmembrane segments in the proteins. The secondary structure of pig PCYT1A was mainly made by α helixes, and β sheets. CCT like superfamily typical domains were found in pig PCYTIA. The protein of PEMT is a hydrophobic protein and there wasn’t a strongly signal peptide, but contained3transmembrane segments. The secondary structure of PEMT was mainly made up of α helixes, disorder coils, and a few β sheets. PEMT like superfamily typical domains were found in pig PEMT. Homology analysis showed that relationship between Sus and Bos aurus was closest, indicating that these two genes were relatively conservative between Sus and Bos aurus in evolutionary development.2Analysis of mRNA expression of PCYTIA and PEMT gene in different tissues and in muscles of Suhuai pig.RT-PCR was employed to examine the relative expression level of PCYT1A and PEMT gene in different tissues. The results showed that the relative expression level of PCYT1A in kidney as well as that in liver were significantly higher than that of heart (p<0.01). The relative expression level of PCYT1A in leaf fat was significantly higher than that in longissimus dorsi (p<0.01), and lower than that in heart (p<0.05). Expression level of PCYT1A in longissimus dorsi was higher than that in musculus psoas major (p<0.05), and was lower than that in complexus (p<0.05). Difference between complexus and musculus psoas major was extremely remarkable (p<0.01). The relative expression level of PEMT gene in kidney was significantly higher than that in heart (p<0.01), and was significantly lower than that in liver (p<0.01). The relative expression level of PEMT in heart was higher than that in longissimus dorsi(p<0.05), with no difference with that in leaf fat (p>0.05). The relative expression level of PEMT gene in longissimus dorsi and complexus were higher than that in gluteus medius (p<0.05), with no difference with that in complexus (p>0.05).3PC content detection in different tissues and muscles of Suhuai pig.PC content in different tissues was detected by HPLC. The results showed that PC content of liver was significantly higher than that of heart (p<0.01). PC content of longissimus dorsi was significantly higher than that of leaf fat (p<0.01), and lower than that of heart (p<0.01). But there wasn’t difference between kidney and leaf fat with the lowest that of kidney. PC content of complexus was higher than that of longissimus dorsi (p<0.05). There are three layers muscles in streak. PC content of the third layer was higher than that of the second layer (p<0.05), and the latter was significantly higher than that of the first layer (p<0.01).
