Western flower thrips Frankliniella occidentalis (Pergande), firstly found in Beijing in the mainland of China, in2003, is a cosmopolitan agricultural pest and one of the most important quarantine pests in the world. With small body type and strong concealed, F. occidentalis also have special mode of reproduction which made the resistance on the rapid development which is difficult to control. Western flower thrips were screened by the glass-vial plus leaf dipping method in laboratory. Using iTRAQ techology coupled with LC-MS/MS, differential proteomic were analysised between sensitive strains of Frankliniella occidentalis and resistant strains of Frankliniella occidentalis. Based on the cytochromeP450gene of mtDNA in Frankliniella occidentalis, cytochromeP450was cloned and heterogenous expressed. At the same time using thermal fogger, west flower thrips were prevented in fogging concentrate.All results showed as following:1. By the drug membrane plus leaf dipping method, Frankliniella occidentalis were screened by chlorpyrifos in laboratory, and determined the sensitive level of F. occidentalis by chlorpyrifos toxicity regression line. LC75corresponding chlorpyrifos was uesed to screen2th instars of Frankliniella occidentalis. When these instars builted a certain scale groups, repeat the above process, when screened19times, sensitivity of Frankliniella occidentalis was decreased39times.2. Sensitive strains of Frankliniella occidentalis is the control group.Collecting300heads Frankliniella occidentalis from both strains, the total protein of sensitive strains of Frankliniella occidentalis or resistant strains was extracted and labeled by isobaric tags for relative and absolute quantitation (iTRAQ). The labeled proteins were isolated by liquid chromatography tandem mass spectrometry (LC-MS) and identified by mass spectrometry (MS). Results:773proteins were identified, among which20were different expressed. These differential proteins can be functionally categorised into binding, catalytic activity, transporter activity and enzyme regulator activity, as well as involving in biological processes of metabolic process, developmental process, biological regulation and response to stimulus. Most of these poteins were in cell part,organelle and macromolecular complex, few was in extracellular region. These differential proteins also participated in various pathways, for example,metabolic pathways,biosynthesis of secondary metabolites, oxidative phosphorylation. Compared with the NCBI, primers were designed, and cytochromeP450gene of mtDNA in Frankliniella occidentalis was selected and cloned, then this gene was heterogenous expressed.3. In the study, the control effect of thermal fogger on western flower thrips was determined in laboratory with different ratio hot fogging concentrate, different solvent, and different spray methods.The results showed that the control effect with fogging concentrate is higher than the control effect with water agent, the highest control effect could rench95%. In addition, Mixing ratio by1:3:6with pesticide:solvent:water, the droplet well distributed and had large quantity in one visual field, which was beneficial to diffuse.
