The western flower thrips,Frankliniella occidentalis(Thysanoptera: Thripidae),has became a great harmful invasive pest in recent years.The ecological and reproductive abilities of the invasive pests are strong.Once the invasion succeed,its propagation and breeding trend are difficult to be controlled.Eventually it will causes serious damage to the ecological environment,agricultural,forestry,and human body health.The western flower thrips is tiny and the early symptoms of infection is concealed,the number of the host plants is numerous,the ability of survival and reproduction is strong.Due to these features,the prevention and control work cannot be carried out quickly and effectively.It also causes great damage to agricultural production.Therefore,finding an effective measure to prevent the western flower thrips has became an urgent challenge.This study uses molecular biology and proteomics research methods to analyze the fitness related proteins of the western flower thrips.Tries to reveal the high invasive power of the western flower thrips on the protein level,so as to provide a new idea to pest risk analysis of the western flower thrips,achieve pest control and increase crop yield.Protein sample preparation is the prerequisite of obtaining an optimal Two-dimensional Electrophoresis(2-DE)gel map.Establishing a suitable 2-DE System can provide a strong support for the subsequent research.In this study,the protein extraction methods(Tris-phenol,TCA/acetone precipitation and Direct decomposition extraction method)of the western flower thrips were analyzed,and the 2-DE system was optimized.According to the extraction rate of protein,the result of SDS-PAGE,2-DE image quality and spot numbers,the results indicate that the TCA/acetone method can remove interfering substance effectively.It is optimal for protein extraction of F.Occidentalis.After the optimal protein extraction method was established,the differential proteomics technology was used to analyze the differential proteins in the westernflower thrips and the flower thrips(Frankliniella intonsa).A 2-DE gel map was got.The protein spots of the difference expression were analyzed by the Image Master2 D Platinum 6.0 software.There were 55 differential protein spots with protein abundance of more than 2 times and good repeatability.We choose 31 of up-regulated expression protein spots and the MALDI-TOF/TOF MS analysis was performed.Hence,20 differentially expressed proteins were identified successfully.Including Inorganic pyrophosphatase ? Mitogen-activated protein kinase-binding protein 1?Heat shock protein?Small heat shock protein 4?Protein lipidation?triosephosphate isomerase?Glutathione S-transferase?Arginine kinase?Calponin and so on.On the basis of NCBI_Thysanoptera database and Uniprot_insecta database,related bioinformatics analysis were done to analyze the function of the differential expression proteins.The results revealed that the proteins' biological processes were mainly involved in abiotic stress process,immune response process,embryonic development process,cell repair process,protein folding process,drug resistance process,stress response process,oxidation resistance process,cell movement process,cell division process,cell signaling process,actin cytoskeleton process,energy metabolism process,and so on.Using Cytoscape v3.4.0 software,a network of 13 adaptive proteins was established and 5 interactions were displayed.Among these interactions,Co-expression,Physical interactions and Genetic interactions occupied a fairly large proportion of 35.37%,34.43% and 24.48% respectively.Co-localization and Shared protein domains represent a small part of these interrelationships.Blw is at the center,the number of links connected with other genes is 16.The results of this study offer the theoretical foundation for the study of revealing the ecological adaptability in alien invasive species Frankliniella occidentalis.This paper provides a new idea to explore new ways of pest control.
