In this study,’pinguoli’ and brown peel bud sport (’yanguangli’) were used as experiment material, paraffin section and PCR amplification technology were used to compare anatomical structure, genome and transcriptome of two pears variety (strain). Determine period for phellem forming of’yanguangli’. Phellem forming is related to lignin synthesis. Find out difference for genome and transcriptome of two pears variety (strain). The main results are as follows:When bad factors stimulated stomatal of rupture, the phellogen formed phellem and lead to ’yanguangli’ peel turn brown. Key period of phellem forming is42nd day after anthesis to56th day after anthesis.The SSR analysis, the forming mechanism of Brown peel of ’yanguangli’ is different with hybrid progeny of ’huangjin’ and ’dangshansu’In DNA, there are462bands were produced from ’pingguoli’ and’yanguangli’ by154primers. In mere5primers (ME4-EM6、ME7-EM5、ME10-EM4、ME14-EM7、 ME14-EM8) to detected7different bands, percentage of polymorphic bands was1.52%.In cDNA of49th day after anthesis peel, there are444bands were produced from ’pingguoli’ and ’yanguangli’ by154primers. In mere2primers (ME4-EM3、 ME7-EM5) to detected2different bands, percentage of polymorphic bands was0.45%.The ME4-EM3primer combination is lectin by Blast, after verification, the different band fromation by the different primer binding sites of ’pingguoli’ and’yanguangli’Brown peel of’yanguangli’is related to anthocyanin and lignin accumulate.
