According to research, The fiber color of brown cotton had close relationship withlight, but it was unclear whether the process of fiber pigment synthesis were related to theregulation of light quality, The relationship between fiber pigment synthesis, accumulationand light needed further study. Therefore, this experiment regards the variety of Bai quanbrown cotton as the research object, and initially established an ovule culture system ofbrown cotton.The condensed tannin content at different developmental stages, fiber lengthwere measured; Using cDNA-SRAP technology, the gene expression of different lightquality treatment in vitro cultured brown cotton fiber were analysised. Moreover, Theinfluence of light quality on fiber development-related enzyme activity and the condensedtannin content were investigated.The main results are as following:(1) Based on BT medium, added the appropriate hormone combinations:1mg L-1GA3+1mg L-1NAA, initially established the Ovule Culture System of BrownCotton and white cotton. With the prolonged incubation time, the fiber length of browncotton and white cotton showed a gradually increasing tendency. Compared with whitecotton, brown cotton fiber length is always lower than the white cotton. In the fiber culture5-20DAC period, soluble proanthocyanidins and insoluble proanthocyanidins firstlyincreased and then decreased gradually; The insoluble proanthocyanidins was alwayshigher than the soluble proanthocyanidins.(2) The light quality treatment can affect the development of fiber in brown cotton.Compared with the control, The sucrose synthase and beta-1,3-glucanase activity ofbrown cotton fiber treated by different light quality were significantly lower. Light qualitytreatment mad both soluble and insoluble proanthocyanidins reduced, as follows: control>white> red> yellow> blue-violet light treatment. The soluble proanthocyanidins werehigher than the insoluble proanthocyanidins in all treatment.(3) Differential expression analysis of the effect of light quality on tissue culture of browncotton fiber by cDNA-SRAP. Screened from80primer combinations,28reproducible,clear and rich polymorphism primer combinations were achieved, These primercombinations produced a total of401clear bands, each primer combinations had average14bands, Among them,58were polymorphic bands,14.46%of the total number of bands,each primer pair generated2.07polymorphic bands. Differential fragment sequencing,homology comparison and functional analysis showed that:16transcription derivative fragments had the homologous with known genes, including the hypothetical protein(Gb-5, GB-10), functional genomics (Gb-1, Gb-3, Gb-7, Gb-8, Gb-13, Gb-14, Gb-15,Gb-16), the chloroplast gene (Gb-11), proanthocyanidins synthetic genes (Gb-4of5),genes about cotton fibers development (Gb-2Gb-6) and resistance gene (Gb-Gb-12).
