Our study is about effect and repair of Polysaccharide from rape pollen on the acute chemical liver injury in mice, to discuss effect mechanism of Polysaccharide from rape pollen.Ultrasonic-assisted extraction was adopted to extract polysaccharide from ultra-fine powdered rape pollen. Water extract-alcohol precipitation was used to extract polysaccharide from rape pollen. The content of polysaccharide was determined by the spectrophotometer using sulphuric acid phenol method. The optimal conditions were determined with the orthogonal design, to obtain the best extraction process conditions is the ratio of solid to liquid1:4, time of60min,temperature of80℃and power of300W, and the rate of extraction is5.956%.This experiment adopts120mice, randomly divided into two groups as "a"、"b",60in each group. Then group "a" is divided into six groups at random, namely, normal group a1, model group a2, the low dose group a3(100mg·kg-1),the middle dose group a4(200mg·kg-1), the high dose group a5(400mg·kg-1), biphenyl diester positive control group a6(150mg·kg-1).As group b too. Except the normal group and model group, Corresponding drugs were to fill the stomach on other groups. Except the normal group,0.1%CCl4Peanut oil was injected to other groups. Kill the mice of a groups after24hours, and kill the mice of b groups2weeks later. Firstly, liver and spleen index、the activity of serum aminotransferas、the content of total bilimbln (TBiL)、the activity of NAGase、antioxidation index of serum、the content of nitrogen monoxidum (NO) and the activity of nitric-oxide synthase(NOS) were detected. Then the microstructure changes of liver tissue were observed. Through all the experiments above, the effects of polysaccharide from rape pollen on liver injury mice were studied systematically.(1) The mice were administrated orally with polysaccharide from rape pollen two weeks, the rape pollen polysaccharide groups could decrease liver index, and decrease the content of ALT、AST and TBiL in serum, also decrease the activity of NAGase in serum. The mice were administrated orally with polysaccharide from rape pollen four weeks, the effects of each rape pollen polysaccharide dose group were all obviously.(2) The mice were administrated orally with polysaccharide from rape pollen two weeks, rape pollen polysaccharide could increase the activity of SOD in serum and decrease the content of MDA in liver homogenate. and increase the content of NO and the activity of NOS, The mice were administrated orally with polysaccharide from rape pollen four weeks, the effects of each rape pollen polysaccharide dose group were all obviously.(3) The mice were administrated orally with polysaccharide from rape pollen two weeks, Microstructure of liver of tested mice were observed which showed that lobular contour, disorganized hepatocyte and hepatic tissue in normal structure is destroyed in CCl4model group, wide moderate and severe hydropic degeneration or ballooning degeneration and fatty degeneration, Nucleus disappears or pyknosis and red-dyed. The safeguard effects of each rape pollen polysaccharide dose group were all obviously. The high dose group was best of all. The mice were administrated orally with rape pollen polysaccharide four weeks, the repair effects of each rape pollen polysaccharide dose group were all followed obviously.
