According to the research of Structure and function of Cordyceps polysaccharide on carbon tetrachloride-induced acute liver injury in mice, Study on the mechanism of Cordyceps polysaccharide, for further research of Cordyceps polysaccharide on animal health, particularly in terms of liver application and promotion of scientific information and provide a theoretical basis.This test uses water extraction and alcohol precipitation method to improve the extracting processt of Cordyceps polysaccharides. Extraction by orthogonal experiment L9(34) to optimize the conditions of Cordyceps polysaccharide, phenol-sulfuric acid method detemine polysac-charide content, the best method of optimum conditions for the extraction process and the extraction rate:extraction time 3 h, feed temperature 80℃, solid-liquid ratio 1:30, extraction rate was 17.14%.Experiment design and implementation. All 120 males laboratory mice were fed one week after adaptation, were randomly divided into the experimental group A and group B. A and B group are located control group, CC14 model group,Cordyceps polysaccharide low-dose group 70 mg·(kg.d)-1, middle dose 140 mg·(kg.d)-1,high-dose group 280 mg·(kg.d)-1,DDB group 140 mg·(kg.d)-1,10 mice per group. In addition to the control group and model group, the other groups are in the appropriate drug (by 0.2mL·20g-1) by gavage. In the given medicines the first 8 d, first 11 d and 14 d, except control group, each test mouse are gavage (0.2mL·20g-1) of 0.2% CCl4 peanut oil. A group at 14 d after CCl4 peanut oil gavage 16 h later, all the mice tested eye off the neck and bled to death, anatomized and drawing materials respectively. Group B continued to gavage two weeks, then eye off the neck and bled to death, anatomized and drawing materials respectively. Sera tested in mice have:AST, ALT, ALP, total bilirubin, and N-acetyl-β-D-glucosaminidase (NAGase); liver homogenates has determined:SOD,GSH,MDA; liver observation and liver tissue apoptosis detection of tissue microstructure, more systematic study about the structure and function of the North Cordyceps polysaccharides on CCl4 induced acute liver injury on mice, to provide a scientific basis of the protect functionon liver of Cordyceps polysaccharide, health and veterinary Clinical and animal breeding on the application.The results of study are as follows:(1) The effect of Cordyceps Polysaccharide on CCl4 induced acute liver injury in mice about the serum indexes ALT, AST, ALP and serum total bilirubin and NAGase. After fed Cordyceps polysaccharide two weeks, compared with the model group, Cordyceps polysaccharide dose group can decrease the serum indexes ALT, AST, ALP activity and serum total bilirub in each test group of mice, NAGase activity decreased, the high dose group and DDB group were extremely significant difference (P<0.01). Cordyceps polysaccharide gavage four weeks, compared with the control group, the serum indexes ALT,AST, ALP,NAGase activity of Cordyceps polysaccharide high dose group, the difference was not significant (P>0.05), serum total bilirubin levels were significantly different (P<0.05).(2) The effect of Cordyceps Polysaccharide on CCl4 induced acute liver injury in mice about homogenates indexes SOD,GSH,MDA. After gavage Cordyceps polysaccharide polysaccharide two weeks, compared with the model group, Cordyceps polysaccharide can increased the activity of SOD and the content of GSH, decreased the content of MDA in each test group of mice liver homogenates, the effect of Cordyceps polysaccharide low dose group were significantly different (P<0.05), high dose group was significantly (P<0.01). After gavage Cordyceps polysaccharide polysaccharide four weeks, compared with the control group, each dose group of Cordyceps polysaccharidehave obvious effect, including high dose group was best,compared with the control group SOD activity was significantly increased, GSH,MDA content had no significant difference (P>0.05).(3) Optical microscopy observed (HE Dyeing). The liver structure of model group was severely damaged, coagulation necrosis of hepatocytes around the central vein, the liver cells showed a wide range of moderate to severe blisters degeneration or degeneration and fatty degeneration of the liver cells into large unstructured red dye and cell debris, a small amount of residual hepatocyte cell degeneration in varying degrees. After gavage Cordyceps polysaccharide two weeks, Cordyceps polysaccharide each test group showed a certain resistance to injury and repair. High dose group liver cells arranged in more orderly, some mild swelling of the liver cells, cell death was significantly reduced, increased dualcore cells, showing the repair process more significant liver injury. After gavage Cordyceps polysaccharide four weeks,each test group were seen in liver tissue degeneration and fatty degeneration of significantly reduced or even disappear, only a small amount of necrosis, and Cordyceps polysaccharide high dose group and DDB Group hepatocytes dualcore cells increased.(4) The effect of Cordyceps polysaccharide on cell apoptosis in CCl4 induced acute liver injury mice. After fed Cordyceps polysaccharide two weeks, light microscopy, the model group showed a larger number of liver cell apoptosis, single or in groups distributed; Cordyceps polysaccharide group have small number of apoptotic cells, scattered. After fed Cordyceps polysaccharide four weeks, the number of apoptotic cells was significantly reduced in the model group, the number of Cordyceps polysaccharide group liver cell apoptosis was significantly reduced, where the most obvious effect of high dose group, showed that Cordyceps polysaccharides on liver of acute liver injury in mice repair.Innovations are as follows:(1) Made a systematic study about the effect of Cordyceps polysaccharide on the structure and function of acute liver injury in mice.(2) Made a systematic study about the effect of Cordyceps polysaccharide on cell apoptosis of acute liver injury in mice.(3) To investigate the correlation Cordyceps polysaccharide between serum NAGase in CCl4 induced acute liver injury in mice. To provide a reference for NAGase application inveterinary clinical application.
