Flue-cured tobacco is important economic crop in China. It requires multiple generations and years to produce varieties which possess parents’ good traits and genetic stability by traditional cross-breeding methods. However, Dihapiloid hybrids from traditional cross-breeding will shorten the breeding process. This study used three hybrids varieties, to get flue-cured tobacco haploid by2ways:unpollinated ovary culture and hybriding with wild tobacco N.africana. The main results of this research were as fellows:1. The period of ovary development has influence on the frequency of embryos induction, the earlier to be planted and the better to be cultured. Coker371Gold×Longjiang911has the highest frequency of embryos induction at the time when petals is going to expose the calyx. Studies on the comparison of different sterilization methods showed that the highest frequency of embryos induction (90%) was produced using,0.1%Hg2Cl treated whole flowers, then gridiled the ovary and incubated the ovary at an appropriate Growth of ovule was produced on the medium with different hormone regimes,160%inorganic salts based on H medium supplemented with the hormone regimes0.5mg/L IAA+lmg/L KT andlmg/L2,4-D+2ml/L6-BA were the best for haploid inducing. Moreover, the vitrification can be can effectively curbed in the medium with9g/L agar.2. Using the wild tobacco N.Africana induced parthenogenesis of3flue-cured tobacco F1hybrids to harvested haploid. After sowing, seedling development to the small cross some dead, some survive. Randomly chose8individuals of Yunyan87×Jiyan9,A1-A8then do flow cytometry to them. The results showed that the A1-A8are not haploid.
