The feed enzyme applied in the animal feed has become an important item in the science and technology revolution of feed stuff in the21century. The main aim of this study was to investigate the effects of four monomeric enzymes preparations supplemented to wheat-based diets on the digestibility of non-starch polysaccharides (NSPs) and performance of broiler chickens by pre-digestion method. The four monomeric enzymes included xylanase, P-glucanase, celluase and mannanase. The results are as follows:1) The experiment was designed to study the equivalent relationship between non-starch polysaccharides monomer enzyme and the amount of reducing sugar, and then to determine the best enzyme preparation, as well as the process parameters, including moisture, temperature and time, on the basis of measuring reducing sugar content. The parameter of the treatment for external predigestion was calculated based on the current investigationand research:the moisture of feed of30%, treat temperature of50℃, and the treattime of60minutes.2) The study was conducted to explore the effect of different combinations of enzyme preparation on enzymatic hydrolysis of broiler diets, and to optimize the best enzyme preparations combination and interaction effects, in the case of the best pre-digestion processing parameters. Combined with the data analysis, the best combination of enzyme preparation of the experimental diets:xylanase186mg/kg, β-glucanase440mg/kg,cellulose337mg/kg, mannanase200mg/kg (P<0.01). The interaction effect on the diets reducing sugar of four monomer enzyme was xylanase> cellulase>β-glucanase> mannanase.3) The aim of this stuty was to make sure the pobility of take indicator method instead of total feces collection methodjust using the design that determining the crude fiber digestibility in broiler diets. The results indicated that the measured valu of4N-AIA was the most high, total feces collection was the second, and the least was TiO2.Fecal recovery of TiO2averaged97.5%, whereas that of4N-AIA averaged103.6%based on total fecal collections. Correlation analysis showed that there were obvious correlation between the valu of crude fiber digestibility determined by total fecal collection method,4N-AIA method and TiO2method(P<0.01), the coefficient were0.968,0.972and0.972respectively. Indicator method could be used to replace total fecal collection method in determining crude fiber digestibility for broiler chickens.4) After the treatment of predigestion, the feed with enzyme had significant effect on growth performance and nutrient digestibilily. Compared with ordinary method of using enzyme in broiler chicken, the dry matter increased8.53%(P<0.01), crude protein16.27(P<0.01), crude fber40.89%(P<0.01), Ca12.17%(P<0.01), P80.99%(P<0.01); considering the indicators of growth performance, there were significant improvements on the feed intake, weight gain and whole F:M for broiler chickens.
