Wheat stripe rust is one of the most important diseases wheat production in China, it occurrence area is very wide, epidemic, high hazard rate, and makes very serious harm. Comprehensive prevention that breeding disease-resistant wheat cultivars is the main way to control wheat stripe rust disease. However, due to the small volume of wheat stripe rust, rapid reproduction, population, toxicity variation of high frequency, new physiological small kinds appeared and became the dominant species, often resulting in the "lost" rust resistance of large varieties in wide area, leading to disease epidemic. Therefore, in order to prevent the occurrence of wheat stripe rust new virulence genes give harm to the production of wheat, to monitor and predict the physiological races of wheat stripe rust became an important link in the prevention and control of wheat stripe rust, wheat breeding of resistant cultivars and distribution.In2010, researchers have found a new stripe rust races V26that can overcome the stripe rust resistance gene Yr26, which can overcome the gene Yr26that is contained in the China’s new breed of a large number of fine varieties of wheat,it again brought serious threat to prevention and control of wheat stripe rust. At the same time,in the2001-2006wheat stripe rust samples that were collected from Taibai County Baoji City, Shaanxi province can be infected with "four" of wheat stripe rust new strains of T4, is currently the only can infect "strains in four." However,"four" variety is widely used in breeding for rust resistance, large area popularization and application and the background of the new resistance source of rust resistance varieties will be more and more in our country in the future. Therefore, research on new strains of T4has become a pressing matter of the moment.In recent years, the molecular biology provides us new way to monitor the constitution and change of PST races as its development. The PST is an obligate fungus, the sexual time is not been foud. In order to establish a practical molecular identification system of PST,specific molecular markers of the prevailing races in China were searched on a large scale.The results were obtained as follows:1. For design RAPD system of PST, the RAPD system of PST was developed. The manyexperiments indicated that the system is more steady and its Repeatability is good.2. In this study, Through seven main physiologic races of Puccinia striiformis f. sp. tritici (CYR29、CYR31、CYR32、CYR33、T4、Sull-4、 V26) were analysed with189primers by RAPD technique.The special fragment of V26new strains was obtained by the primers S1390and S1325.The special fragment of T4was obtained by the primers S1311and S1363.3. Because the special fragment The special fragment of V26new strains was obtained by the primers S1390are stable.So wo recovered, purified, sifted and clonedthe special fragment. Based on the sequencing result, specific PCR primers were designed and a SCAR marker for V26new strains was obtained successfully by detecting seven main physiologic races of Puccinia striiformis f. sp. tritici.4. For the new strains T4that can infect the wheat,now we only amplified out two specific bands, we analyze its sequence and design a few specific primers after cloning and sequencing, But due to various factors, SCAR marker monitoring system has not been successfully transform.
