Geminiviruses are a group of single-stranded circular DNA plant viruses, which occurred world wide, and caused severe diseases in several economically important crops.Several kinds of begomoviruses were identified in tobacco, tomato, squash,et al in China since1990s, begomoviruses infecting Lycopersicon esculentum var.cerasiforme and Cucumis melo var. Saccharinus were identified in this dissertation.Thirty-two virus samples collected from Ledong and Sanya were conformed to be infected by geminiviruses by PCR detection with PA/PB. WTG were detection from23samples. The complete nucleotide sequence of DNA-A-like molecule of FQ12contained2774nucleotides. FQ12DNA-A had the highest sequence identity (81.9%) with that of Malvastrum leaf curl virus isolate G100. Based on the molecular data, FQ12was considered as a new Begomovirus species, for which the name Tomato leaf curl Hainan virus, ToLCHNV was proposed. FQ13DNA-A had the highest sequence identity (96%) with that of Ageratum yellow vein China virus-[Hn2-19], Based on the molecular data, FQ13was an isolated of AYVCNV(AYVCNV-[FQ13]). Satellite DNA-β with conserved primer pairs by PCR had been unsuccessful. Co-infections of begomoviruses exist on Lycopersicon esculentum var.cerasiforme.Twelev virus samples collected from Ledong were conformed to be infected by geminiviruses by PCR detection with PA/PB. WTG were detection from5samples. The complete nucleotide sequence of DNA-A-like molecule of HA1and HA3contained2736nucleotides. HA1、HA3DNA-A had the highest sequence identity (97%and99%) with that of Squash leaf curl China virus isolate Hn61(SLCCNV-[Hn61]). HA1、HA3were isolated of SLCCNV(SLCCNV-[HA1], SLCCNV-[HA3]). Satellite DNA-β with conserved primer pairs by PCR had been unsuccessful.
