Dendranthema×gradifolium(Ramat) Kitam, which belongs to Chrysanthemum, Compositae,originated in China and the germplasm resource is very abundant. It is one of the ten traditional famousflowers in China. There are30species in all, and17species in China. At present, there are more than tenthousand kinds of Chrysanthemum cultivars in the world, and more than three thousand kinds in ourcountry. Because of existing lots of cultivars and variations, and the work of its background andphylogenetic relationship was greatly restricted, which limits the process of genetic improvement and theapplication of landscape practice for Chrysanthemum.AFLP (amplified fragment length polymorphism) is a new DNA molecular marker technique basedPCR with good reliability, good repeatability, high reliability and rich polymorphism and individualspecificity. It has been widely used in classification, cultivar identification, genetic mapping, genetic andbreeding. Seventy Chrysanthemum cultivars were analyzed. Six pairs of primers were chosen from64pairsof primers with AFLP analysis and generated191bands in total, in which181bands were polymorphic.The percentage of polymorphic bands was94.8%, with an average of30.2bands produced per primer pair.Cluster analyses by UPGMA showed that there were close genetic relationships among cultivars of thesame petal type.SSR(simple sequence repeat) is a DNA molecular marker technique based on DNA lengthpolymorphism, and it is a very useful tool for the analysis of population genetic structure and geneticlinkage map construction. A total of8microsatellite markers were developed to study population geneticsof Chrysanthemum using the Fast Isolation by AFLP of Sequences Containing Repeats technique. Six ofthe markers were polymorphic in a sample of44individuals obtained from five populations in China.
