Analysed Polymorphisms Of Growth Axis Related Genes Associate With Growth Traits In Poll Dorset Sheep

DNA sequencing techniques was applied to analyze the genetic variation of GH gene (exonl&exon4), GHR gene (exon10), IGF1gene (exon5), IGF2gene (exon3) and Myf5gene (exon1&3) with160Poll Dorset sheep from XinAo sheep farm of Manas Xinjiang in this experiment. And measured growth traits from one month to seven month, include birth weight, body weight, body height, body length, shoulder width, hip width, chest circumferences and chest depth. Using least square method analyzed the relationship with the SNP site and growth traits in Poll Dorset sheep, the results showed.There were no single-nucleotide polymorphisms mutation of the1st and4th exon locus on GH gene, the5th exon locus on IGF1, the3rd exon locus on IGF2gene and the1st and3rd exon on Myf5gene in the Poll Dorset.Two SNP mutations were found in the10th exon of GHR gene. The first SNP site located on634locus, three genotype AA, AG, GG existed in the group, frequencies of genotype are0.0948(AA),0.5603(AG),0.3449(GG), frequencies of allele genes A/G are0.3750/0.6250, G is the dominant allele. The mutations of amino acids by aspartic acid (D) into asparagine (N). Analysed the correlation with genotype and growth traits, the results showed, at the two-month-age, AA and AG had the most significant difference in the body weight (p<0.01), AA also had significant difference with GG (p<0.05), the average body weight of the three genotype is AA<GG<AG. AA and AG, AA and GG had significant difference in the body height (p<0.05), the average body height of the three genotype is AA<GG<AG. In the three-month-age, AA and AG, AA and GG were significant difference in the chest circumferences (p<0.05), the average chest circumferences of three genotype is AA<AG<GG. In the two and three months’age, AA and AG, AA and GG were significant difference in the hip width (p<0.05), the average hip width of the three genotypes is AA<GG<AG. AA, AG, GG genotypes had no significant difference in the body length, chest depth and shoulder width (p>0.05).The second SNP site located on804locus, three genotype AA, AG, GG existed in the group, frequences of genotype are0.6983(AA),0.2845(AC),0.0172(CC), frequences of allele genes A/C are0.8405/0.1595. A is the dominant allele.The mutations of amino acids got from glutamic acid (E) to aspartic acid(D). Analyzed the correlation with genotype and growth traits, the results showed:at the two-month-age, AA and AC were significant difference in the shoulder width (p<0.05); the average shoulder width of three genotype is AC<AA<CC. At the three-month-age, AA and AC were significant difference in the body height (p<0.05); the average body height of three genotypes is CC<AC<AA. AA and AC were significant difference in the chest circumferences (p<0.05); the average chest circumferences of three genotype is AC<CC<AA. At one-month, two-month, four-month, and seven-month-age, AA and AC were significant differences in the hip width (p<0.05), At one-month-age, the average hip width of three genotypes is AC<AA<CC, at two-month-age,the average hip width of three genotypes is CC<AC<AA, at four-month-age and seven-month-age, the average hip width of three genotypes is AC<CC<AA. AA, AC, CC genotypes had no significant differences in the body weight, body length and chest depth (p>0.05).In conclusion, analysis relationship with SNP site located in634locus and growth traits,its indicated that AG and GG increase body weight and body height at two-month-age, the hip width, chest circumferences also expand more at three-month-age than AA, G is the dominant allele which interfered with those traits of the Poll Dorset. Analysis the SNP site located in8041ocus and growth traits, it showed that the growth traits of homozygote (AA and CC) were higher than heterozygous (AC), which SNPs effects on growth traits undetermined in these Poll Dorset.
