| Mycoplasma bovis(M.bovis) is an important cause of pneumonia and arthritis in young calves, and mastitis. In recent years, the cattle disease caused by M, bovis has spread widely to all parts of the world. Since2010, in some areas of the North of Xinjiang large-scale dairy calves successively broke out severe pneumonia and arthritis that resulted in the deaths of more than10calves.The laboratory diagnosis confirmed pathogen was Mycoplasma bovis. At present, there were no vaccines for prevention and control of Mycoplasma bovis disease in our country, once outbreaks of disease relying mainly on antibiotic treatment. But now there are wide variety of antibiotic, blind medication and bad treatment. In order to solve the above problems, the experiment has focused on the following research:1. To evaluate the immunoenhancement of complete freund’s adjuvant, oil—emulsion, aluminum hydroxide and propolis on inactivated vaccine against Mycoplasma bovis disease, we combined the four adjuvant and Xinjiang local isolated strains to prepare inactivated vaccine, and immunized healthy mice respectively, ELISA method was used to detect the serum antibody titer. The results showed that the serum antibody titer of oil—emulsion group was highest and longer duration, which was the best adjuvant.2. On the basis of Cattle Heart Broth Medium, cattle heart broth was concentrated two times and also increased DMEM and cattle lung broth, the growth titers of Xinjiang local isolated strains in the improved Cattle Heart Broth Medium were determined by viable count method, bacterial protein determination method, OD value determination method and plate count method respectively. The results showed that the efficiency of Mycoplasma bovis culture in improved medium was significantly improved, which was helpful for mass culture.3. The minimal inhibitory concentrations(MIC) of enrofloxacin, ciprofloxacin lactate, lincomycin hydrochloride, tylosin, azithromycin, tilmicosin, ofloxacin and florfeniol on Mycoplasma bovis in vitro were determined by two times dilution method. The results showed that resistance to enrofloxacin was minimum, which was applicable to the current treatment of Mycoplasma bovis infection diseases. |
