The plant area and yield of castor in China is larger than other countries. Most varieties of castor are tall in height and not uniform in maturity. This results in wasting lots of nutrients and not easy to realize harvesting by mechanized method. The article is designed to locate the genes which are related to regulate stem length, leaf type and yield are screened by modern biotechnology. And it lays the foundation of screening good dwarf castor characterized by internodes short, stout stem body and medium length, high yield.cDNA-AFLP technology combines advantages of RT-PCR and AFLP technology. It has been widely applied in the analysis of specific gene expression, dominant mechanism, functional marker analysis, gene expression characteristics analysis and soon.The generation of dwarf stem castor BC2F2and paternal stem are chose as materials, and the cDNA-AFLP technology is used to analyze the difference in genes expression of stalk at the beginning of flowering period. The differential fragments are recovered, cloned and sequence analysis. Bioinformatics analysis and functional annotation are done at last. This study is expected to discover the associated dwarfing genes in the dwarf castor, find a breakthrough to reveal the molecular mechanism of the dwarf castor. The main findings of this research are as follows:1. The cDNA-AFLP Analysis System for Castor is established successfully.2. The establishment of technology system is used to analyze the differences in gene expression of dwarf strains stem at the beginning of flowering.115primer combinations are used for selective amplification, and458differential bands are received.128target genes which are recovered and cloned are measured by sequence.3. These sequence received are used to do the homology search in NCBI database. The results showed that92genes have high homology compared with the known protein, and the remaining36genes are unknown genes. The proteins expressed by homologous genes are as following. The protein is used for regulating the plant stem elongation or hormone synthesis, regulating cell growth and differentiation in transcription initiation factor, affecting cell growth and development by regulating gene expression or signal conduction, the reaction involved optical and thermal stimulation and oxidation and is also including some growth hormone hydrolase, muscle actin cytoskeletal proteins, transport proteins, and so on.
