| In this study,‘Jintian Huangjia seedless grape’ s selfing embryo was used toconduct morphological observation, to count embryonic abortive rate under naturalconditions,to observe the process of seed development by using slice and to study theinfluence of different conditions on the embryo development. After analyzing the testresults, we can draw the following conclusions:1. Under natural conditions,‘Jintian Huangjia seedless’ grape’s selfing embryobegan to degenerate and a small number of ball-type embryos were still developingafter pollinating65d~75d; after85d~90d pollination, embryo almost disappearedand only the seed with the abnormal and thick exotesta remained.2. At the same time, the exotesta continues to thicken as follows: after50dpollination the epidermis inside cells divided to the center and gradually formed thefence structure. After70d pollination, the inside cells of seed coat divided andelongated to the center, the cells began to expand, cell wall thinned, cell gap increasedand the seed coat succulented. The succulent seed coat’s proanthocyanidins and thecontent of resveratrol is higher than the flesh.3.‘Jintian Huangjia seedless’ selfing embryo and ‘Jintian Huangjia seedless’with ‘Crimson Seedless’ hybrid embryo both have higher development rate in themodified N medium than in the natural state;‘Jintian Huangjia seedless’ selfingembryo and hybrid embryo culture at25℃have higher development rate than in thenatural state and the low temperature treatment does not improve self-rescue successrate of both the selfing embryo and hybrid embryo; Developmental rates is higherwith6-BA1.0mg·l-1+GA30.5mg·l-1than in the natural state; To both selfing embryoand hybrid embryo, saving effect is obvious when adding500mg·l-1LH(lactalbuminhydrolyzate); With sucrose as the carbon source in embryo development,developmental rate is significantly higher than that of lactose and glucose and withlactose as the carbon source, the hybrid embryo’s development is not good. Lowsugar concentration can improve embryo development rate, while high sucroseconcentration inhibit the growth and development of the embryo; The acidicenvironment conducive to the development of both self embryo and hybrid embryo.4. When crossing tetraploid rose fragrance with ‘Jintian Huangjia seedless’, we got low fruit rate and embryo rescue effect is bad, but triploid plants can beobtained. |
