Male sterility in plants is a common phenomenon, In-depth study of male sterility, that can rich cytoplasmic genetic, the pollen developmental and the cytoplasmic and other aspects of theoretical knowledge.Cytoplasmic male sterility in rapeseed breeding is very important and extensive,It can dispense with artificial emasculation of trouble, and to improve the purity of hybrid seeds, which is one of the rape heterosis important way to utilize. Also, because the cytoplasmic male sterility by the nucleoplasm two sets of genetic control of the system, which can be done cytoplasmic the ideal system.In this paper, cytoplasmic male sterile line of Brassica napus09A and the fertile line of09B as a research material from the male sterile line of flower morphology and pollen viability, to the different developmental stages of bud peroxidase (POD), catalase (CAT), superoxide dismutase (SOD), ascorbate peroxidase (APX) on some protective enzymes, and malondialdehyde(MDA) and free proline (Pro) content, then extracting soluble proteins, SDS-PAGE electrophoresis, the difference strip sent to mass spectrometry analysis,the analysis showed that this differential bands is PGK enzyme and PGK gene molecular cloning,in order to reveal the physiological and biochemical and molecular level the genetic mechanisms of male sterility.Brassica napus male sterile line09A in all aspects of floral organ morphology, the protective enzyme as well as malondialdehyde (MDA) and proline content, the dodecyl sodium thiosulfate polyacrylamide gel electrophoresis of protein, and the PGK gene cloning are different from09B.09A flower organs stamens a length shorter than09B, pollen content is much lower than the09B. Four protective enzyme content, malondialdehyde (MDA) and proline have suffered different,09A buds of four protective enzyme activity trends are different from the09B bud, and CAT, SOD, APX activity decreased male sterile active oxygen metabolism may lead to loss of balance, level of lipid per oxidation increased, membrane lipid damage increases, resulting in anther abortion of09A bud.09A MDA content in every stage of bud development is higher than the09B,Proline has an important physiological function of the amino acids, has a very important role in pollen development process,09A lack of proline content appears early in the bud development may be one of the reasons that lead to the09A abortion. Soluble protein electrophoresis showed differences in bands, indicating that the protein content is different.09B and09A to do template then do the PGK gene cloning, sequencing results after return to No.1and NO.2than display, some nucleotide changes. The morphological characteristics of the experiment from09A to the physiological and biochemical characteristics, and then do the PGK gene cloning, the revelation of the full range of multi-level male sterility genetic mechanism, to lay the foundation for a rich theory of male sterility.
