Genetic variations of (KAP6.1、KAP8.1、KAP8.2、KRTAP9-2and KAP16.4)five KAPgenes were detected by PCR-SSCP, and DNA sequencing techniques in418individuals fromChinese Merino Sheep(Xinjiang type), and their genetic structure and diversity were analyzed;meanwhile, association analysis were carried out to detect the genetic effects by using SAS8.1software and Least Square Analysis method on Genetic Polymorphism of five KAP Genes andTheir Associations with Wool Quality Traitsins at the candidate genes of Chinese MerinoSheep(Xinjiang type).The objects were to obtain the molecular genetic information, to find outthe DNA Makersassociated to the Wool Quality Traitsins, and to provide scientific informationto the protection,development and application of in order to provide a theoretical basis for woolsheep molecular breeding:Firstly, Study the genetic polymorphism of5KAP Gene at Chinese Merino Sheep(Xinjiangtype)Choosing5KAP Gene as Candidate gene,Use by primer primer5.0primer design software related Genes were designed6pairs of primers,the Chinese Merino Sheep(Xinjiangtype) were analyzed by PCR—SSCP method.Theresult showed: KRTAP9-2Gene had34bpA/-,35bpC/A,46bpA/-,132bpG/C four mutant site;KAP6.1Gene had54C/T,162G/A,166C/A,167A/T,55bpA/-five mutant site;KAP8.1Gene had53bpC/-,62bp-/T,85C/A three mutant site;KAP8.2Gene had133A/C,46bp-/A two mutant site;KAP16.4-1Gene had89bpG/A,188bpG/A,196bpA/T,190bpA/C,137bpG/A five mutant site;KAP16.4-2Gene had78G/C,35bpA/-two mutant site.The χ2examine showed, KRTAP9-2Gene was at Hardy-Weinberg disequilibrium(P<0.01); KAP6.1,KAP8.1,KAP8.2,KAP16.4-1,KAP16.4-2Gene was atHardy-Weinberg equilibrium(P>0.05).Chinese Merino Sheep(Xinjiang type) was moderate polymorphism at5KAP Gene and PIC was between0.25~0.5.KRTAP9-2,KAP6.1,KAP8. 1,KAP8.2,KAP16.4-1,KAP16.4-2Gene polymorphism information content (PIC) was0.384,0.364,0.484,0.375,0.486,0.351,Genetic variation was midrange level at Chinese Merino Sheep(Xinjiang type).Secondly, study the relation of5KAP gene for wool traitsStudy relation obtained by Genotype phenotypic data least squares mean and standardwool traits (identification weight, wool length score, wool bending score, wool fineness score,wool grease score,wool gloss score, body size, amount of wool,mean diameter, coefficient ofvariation, number of bending)gene loci were analyzed. the result showed that:(1)KRTAP9-2Gene genetic loci amount of wool AA,AB,BB and CC genotypes was significantlydifferent(P<0.05);(2)KAP6.1Gene genetic loci wool fineness score and wool grease score ABand AA,BB genotypes was significantly different(P<0.05);(3)KAP8.1Gene genetic loci woolbending score BB and AB genotypes and wool fineness score AA,AB,BB and CC genotypeswas significantly different(P<0.05);(4)KAP8.2Gene genetic loci amount of wool BB and ABgenotypes was significantly different(P<0.05);(5)KAP16.4-1Gene genetic loci wool bendingscore CC and AB genotypes, amount of wool AB and AB,BB,CC genotypes and coefficient ofvariation CC and AA,AB genotypes was significantly different(P<0.05);(6)KAP16.4-2Genegenetic loci wool grease and wool gloss score AB and AA,BB genotypes was significantlydifferent(P<0.05).other Genotype difference was not significant (P>0.05).Thridly, Study the combined genotypes of the5KAP Genes for wool traitsThe effect of combined genotypes of the candidate genes for wool traits was tested byleast square means in Chinese Merino sheep(Xinjiang type).The results showed that the KAP6.1and KRTAP9-2Genes is more effective wool length score than AABB,wool grease score andwool gioose score than ABAA and ABAB,amount of wool than BBBB and AAAB,number ofbending than ABBB combined genotypes (P<0.05); KAP8.1and KAP8.2Genes is moreeffective identification weight than AACC andABCC,wool bending score thanABAA,AABB,BBBB,BBAB,BBCC and wool fineness score than BBAA,ABAA,ABBB,ABABand body size than AACC,amount of wool than BBBB combined genotypes (P<0.05); KAP16.4-1and KAP16.4-2Genes is more effective identification weight than AAAB,woolbending score than ABAA, ABCC and wool fineness score,wool grease score,wool glossscore,number of bending than ABAB,coefficient of variation than ABCC combined genotypes(P<0.05),and other combined genotype contribution of wool traits was low.
