| In order to study the effect on breast repairion of breast mastitis model rats that after transplantating bone marrow mesenchymal stem cells, BMSCs were injected to rats’breast base, and explore the repaired effects on mammary gland that BMSCs have and possible mechanism of action by detecting the levels of serum inflammatory cytokines TNF-α,IL-1β, IL-2,1L-6,1L-8, immunoglobulin IgM, IgA, IgG, free radical MDA, the activity of antioxidant enzymes SOD, CAT and GSH-Px, enzymes, and observated tissue section,,thus providing a new method for the treatment of mastitis, and laying an theoretical basis on the research for BMSCs repairing mechanism.20SD female rats were selected at early time of the test, they were divided into two groups:experimental group and control group, n=10.10rats were selected randomly for establishing mastitis model by infusing endotoxin through nipple tube, and another10rats were perfused saline in the same way. After24h of Infusing endotoxin, the rats were all killed, mammary gland tissues were collected immediately, fixed for tissue section, cardiac blood was collected from the dead bodies and preserved for detecting the levels of TNF-α,IL-1β,IL-,IL-8,HP,SAA,CRP,MDA,LDH,NAGase. The results showed that TNF-α,IL-1β,IL-6, IL-1, IL-8, HP, SAA, CRP, MDA, LDH, NAGase increased significantly (P<0.05), SOD, GSH-Px decreased significantly (P<0.05); in control group, rats’breast tissues have no obvious changes; after infusing endotoxin,rats in experimental group depressed, anorexia, mammary gland swelled; pathological section showed that mammary gland flocculus become widen, breast tissue collapsed, and were infiltrated by a lot of fallen mammary gland epithelial cells and inflammatory cells, and it proved that mastitis model had been established successfully.After experimental mastitis model was established successfully,90rats were selected and established experimental mastitis model in the above method. After24h of establishing model,90rats were randomly divided into three groups:control group, antibiotic group and BMSCs transplantation group, n=30, and killed5rats each group at test-1,0,1,3,5,7d respectively, mammary gland tissues were collected immediately, fixed for tissue section, cardiac blood was collected from the dead bodies and preserved for detecting serum levels of TNF-α,IL-1β,IL-2,IL-6,IL-8,IgA,IgG,IgM,MDA,SOD,CAT,GSH-PX,NAGase,LDH,MPO.The results showed that, transplanting BMSCs could significantly lower the levels of TNF-α, IL-1β\IL-8at each time point (.P<0.05),and IL-6were significantly higher than control group at1d,3d and5d (P<0.05). The levels of Ig-A, Ig-G, Ig-M at1d,5d,and Ig-G, Ig-M at7d were significantly higher than control group (P<0.05). The level of MDA in serum in BMSCs transplantation group significantly lower than control group and antibiotic group at3d and7d(P<0.05). The activity of SOD in serum in BMSCs transplantation group at1d,3d,5d,7d,GSH-PX at7d, CAT at1d,3d,5d were all significantly higher than control group(P<0.05). The activity of NAGase, MPO in serum in BMSCs transplantation group at7d, LDH at3d,5d,7d were significantly lower than control group(P<0.05).The level of Hp in serum in BMSCs transplantation group at1d,3d, SAA at1d,3d,5d, CRP at3d,7d were significantly lower than control group(P<0.05),there was no different between antibiotic group and BMSCs transplantation group(P>0.05). The results of pathological section showed that, before infusing endotoxin there was no abnormal, acinar epithelial cells arranged in neat rows;24h after injecting endotoxin, mammary acinar structures were damaged severely, partial collapsed, mammary epithelial cell shedding, necrosis, the acinar cavity and interstitial within a large number of exfoliated epithelial cells and neutrophils infiltrating lobular interval widened; breast tissue gradually recovered after treatment, recovered speed of rats mammary in BMSCs transplantation group and the antibiotic group at each time point were significantly more effective than control group. At7day, BMSCs transplantation group had recovered to normal levels.Following conclusions:(Drat mastitis model was successfully established;②BMSCs could reduced the level of TNF-α,IL-1β,IL-8in serum;③BMSCs could increase the level of Ig-M, Ig-A, Ig-G of the immunoglobulin, and enhanced the body’s immune system;④BMSCs could significantly reduce the level of MDA in serum, improve the activities of SOD and CAT in serum, thereby enhance the body’s free radical scavenging capacity;⑤BMSCs could reduce the mastitis serum levels of CRP, SAA, Hp of acute phase protein;⑥BMSCs could reduce the activity of the serum LDH, Nagase, MPO, promote to the repairing of the breast tissue. |
