Objective: It can induce inflammation of mice uteres by tail intravenous injection with LPS, anda mass of macrophages and TNF-α gather in the uteres.Our experiment observe the change of themacrophages and TNF-α by blocking MCP-1to study the effect of MCP-1in mice uteres.Methods:75Kunming mice were randomly divided into3groups, respectively control group(A),Model group (B) and blocking group (C).The mice of group A are handled with tail intravenous injectionwith PBS0.2ml on the first day morning,at the same time of the second day injected with PBS50μl;At thesame time the mice of group B are injected with LPS0.2ml firstly,and the second with PBS50μl;The firsttreatment on group C is the same as the group B,but the second injection is the Polyclonal antibody ofMCP-150μl.The mice of the three groups are put to death after the last injection1h,3h,6h,12h and24hrespectively,and we take out their uteruses.The five time point of group A is marked as A1,A3,A6,A12andA24,the other two groups are also marked like this.The uteruses of the3group are treated with Paraffinsection,we detect the CD14+macrophages and the expression of its CD14,as well as, we detect theexpression of TNF-α and MCP-1which is after blocked by the method of ELISA.Results:(1)The macrophagesof the control group spread over the endomrtrium and perimrtriumof uteres; The number of macrophages of the model group is excessive and uniform distribution; Themacrophages of the blocking group spread over the perimrtrium of uteres.(2) Compared with the controlgroup,the number of macrophages and CD14of the model group increases significantly(P<0.01); theexpression of TNF-α increases significantly(P<0.01)(3) Compared with the control group,the number ofmacrophages and CD14of the blocking group in Endomrtrium and myometrium is in line with A group atthe time point of1h、3h、6h,but increases significantly(P<0.01) at12h and24h. The number ofmacrophages and CD14of perimrtrium is significantly higher than A (P<0.01). the expression of TNF-α isin line with A group at the time point of1h、3h、6h, but increases significantly(P<0.01) at12h and24h.(4)Compared with B, the number of macrophages and CD14of C in Endomrtrium and myometrium dropsignificantly(P<0.01); the number of macrophages and CD14of perimrtrium is obviously low (P<0.01) atthe time point of1h、3h、6h,but they are in line with B at12h and24h. The expression of TNF-α is obviously low (P<0.01).Conclusion:(1)The number of the macrophage increases in the inflammation of mice utereswhich is induce by LPS,and as well as the expression of CD14and TNF-α,this apply to the high expressionof MCP-1maybe.(2)MCP-1play an important role in the distribution of macrophage,it inducemacrophage move from the perimrtrium and myometrium to the Endomrtrium in the uteresinflammation.(3) blocking MCP-1can reduce the expression of CD14and TNF-α,so It can lighten theinflammation of mice uteres.
