Objective: We can conduct in-depth discussions the mechanism of preventing miscarriage ofquercetin by studying the effect of it on mouse’s uterus macrophage,CD14,TNF-α and MCP-1,which areinduced by LPS.This provide both the theoretical basis and scientific on studying new typeimmunosuppressant.Methods: We randomly divide Kunming mice to4groupes: control group(A), LPS abortionmodel group(B), quercetin treatment group(C) and quercetin and LPS double treatment group(D).There are5pregnant rats in each group. From pregnant for two days to seven days, the mice of A and B group arehandled with PBS(0.4ml) by gavage.The mice of A group are injected with PBS(0.2ml) on pregnant for7days,and at the same time the B group are managed with LPS(0.2ml).The mice of C and D group arehandled with quercetin by gavage from pregnant for two days to seven days,then respectively, they areinjected with PBS(0.2ml) and LPS(0.2ml).All the mice are put to death separately at the time of1h,3h,6h,12h and24h after the last treatment.Then we take out their uteruses,observe the morphologe ofembryo, detect the change of CD14+macrophage and the expression of CD14throughimmunohistochemistry and test the variation of the content of TNF-α and MCP-1by ELISA.Results:(1)The rate of abortion of B and D group is significantly higher than A group,but therate of abortion and embryo resorption of D group is significantly lower than B group,the rate of abortionof LPS model group is100%(P<0.01), the rate of abortion of quercetin and LPS double treatment groupfalls to40%(P<0.01).(2) compared with A group,the content of MCP-1and TNF-α of B and D groupincrease significantly(P<0.01)after1h of the last injection.the number of CD14+macrophage increasesignificantly(P<0.01), the expresstion of CD14is up-regulation(P<0.01); compared with B group, thecontent of MCP-1and TNF-α of D group decrease significantly(P<0.01),and the number of CD14+macrophage and CD14as well(P<0.01).(3)The content of MCP-1and TNF-α of B group increase afterafter1h of the last injection,and are still rising at12h and24h. The content of MCP-1and TNF-α of Dgroup reduce after6h of the last injection,and they are close to the expression level of A group,and continuethe downward trend as the time change. Conclusion:(1) The content of MCP-1of the abortion mice uteres increases as the LPS effectingtime went on, and accompanied by the increasing of CD14+macrophage、CD14and TNF-α.(2) Whenuteres inflammation, MCP-1expresses at high level in uterus,it may relate to the removing of macrophagefrom the mesangial area and adventitia to endometrial.Quercetin may weaken migration effects byinhibiting the generate of MCP-1.(3)The effect of quercetin to abortion mice which are induced by LPSmay be by the way of restraining the express of MCP-1,then,adjust the number and distribution ofmacrophage,and then reduce the expresstion of CD14, so as to block the antigen-presenting process ofmacrophage,at last, prevent the excessive expresstion of TNF-α.
