| To explore the the pig genetic variation,in this experiment,17micro satellite loci of the SLA complex region were used, including seven groups of Laiwu pigs, Lantang pigs, Xiaomeishan pigs, Ningxiang pigs, Huainan pigs, Wei pigs and Luchuan pigs; and selected12pairs of micro satellite primers to detect the polymorphisms of F2generation from the Duroc×Erhualian pedigree,and make a association analysis with80-days blood immune parameters,by calculating the number of alleles, heterozygosity and polymorphism information content and other indicators to measure the genetic polymorphism of the SLA groups, and applying the genetic distance and population structure to analysis the population genetic variation, using the R language GLM and immunity to do correlation analysis, the results are as follows:1. We detected genetic polymorphism of17microsatellite loci of228individuals from seven varieties of groups. Totally detected173alleles in the whole population, whole loci of the seven groups are an average of10.18alleles, each loci of the seven groups is an average of4.82.2. We found that three loci polymorphism of SLAMS034, SLAMS045and SLAMS056are low in Laiwu pigs, the loci polymorphisms of SLAMS016, SLAMS034and SLAMS035are low in the Xiao Meishan pigs, and one loci polymorphism of SLAMS056is low,two loci polymorphisms of SLAMS052SLAMS098are low in Huainan Pigs and one polymorphism of SLAMS056is low in the dike pigs, the remaining loci showed good polymorphism. From the the7population level, observed heterozygosity (Ho) is between0.125(SLAMS034) and0.906(SLAMS013), expected heterozygosity (He) is between0.173(SLAMS059) and0.875(SLAMS018), polymorphism information content (PIC) is between0.155(SLAMS059) and0.854(SLAMS018). The SLAMS013and SLAMS098are homozygous in Xiaomeishan pigs.The low polymorphism micro satellite and moderate polymorphism micro satellite are SLAMS045SLAMS056, and SLAMS098(except Lantang pigs) in the five groups; the The highly polymorphic loci are SLAMS005, SLAMS013(exceptXiao-meishanpigs), SLAMS018,SLAMS043,SLAMS052.3. F-statistical results indicated,the genetic differentiation of inter-group is large,22.9%of the genetic variation is caused due to inter-group, and77.1%of genetic variation is caused due to the differences between individuals within each group. We use genetic distance clustering analysis and population structure analysis to analysis the genetic structure of the seven local goat breeds,and the two analysis results are basically consistent. Two analysis methods all divide the seven local pig breeds as following:Wei pigs and Lu Chuan pigs are grouped into one class firstly;secondly, grouped into another class with Huainan pigs and ningxiang pigs for further; Laiwu pigs and Lantang pigs are grouped as a class, Xiaomeishan pigs is clustered in another.4. Detected alleles of12microsatellite loci of392F2generation from the Duroc x Erhualian pedigree, calculated the number of alleles and allele frequencies of each site, got points heterozygosity, and morepolymorphic information content, found them has a more extensive polymorphism within this family groups; in addition, we made a comparison between hybrid pedigrees with local pig to use the polymorphism information better bit of classic gene from near SLA I and II, in the SLA I region, the PIC of local pig is higher (0.537), hybrids (0.496);but in the SLA II,hybrids is higher0.62, local pig is0.53.5. Analysis the relationship of between SLA microsatellite loci and the80-days blood immune parameters of F2generation from the Duroc x Erhualian pedigree. The results show that, in the selectd12microsatellite sites, SLAMS035and80D_blood_parameter (80D)-BA are significantly related (P=0.040), SLAMS034and80D-BA%are significantly related (P=0.026), SLAMS050and80D-PDW are significantly related (P=0.045), SLAMS013and80D-MPV,80D-NE%are significantly related (P=0.025,0.037), SLAMS013and80D-PLT are highly significant correlation (P=0.006), SLAMS056and80D-BA%,80D-HCT,80D-HGB are significantly related (P=0.022,0.011,0.027), SLAMS048and80D-LY%,80D-MCV are significantly associated (P=0.024,0.032),besides, SLAMS048and80D-MHC,80D-WBC are highly significant correlation (P=0.003,0.006), SLAMS01680D-NE was significantly correlated (P=0.031). |
