Abiotic stresses affect plant growth and development, can cause reduction of crop outputor even total crop failure. This reasearch take (Chloroplast Fructose-1,6-bisphosphate aldolase,CpFBA)as the subject. This enzyme is one of the important speed limit enzymes of Calvincycle,and participate in chloroplast of carbon, nitrogen metabolism,responding positively toabiotic stress in some plants. For investigating the biologic function and the expressionpattern of wheat CpFBA in abiotic stresses conditions,studying CpFBA gene how to enhancegenetically modified (gm) wheat resistance as well as how to take advantage of geneticengineering to improve resistance of wheat. In this paper, we took Aibian1, albino line, Chinaspring, Xinong928, Xinong2000and Jinmai47as materials, with semi-quantity RT-PCR, theexpression change of CpFBA in wheat seedlings treated with low temperature, salinity, PEGand abscisic acid were analyzed at transcriptional level. we cultivated and identified T1generation of genetically modified (gm) wheat group which had been successfully transferredCpFBA from our laboratory. in the same way, we identified T2generation of geneticallymodified (gm) wheat group,and measure the difference on soluble saccharide betweenpositive strains and wide type.For another, we cloned wcs120promoter, and replacedconstitutive35S promoter in pU18by WCS120promoter,and then get WCS120-CpFBA-eGFPplant expression vector. The major results as follow:1. the expression levels of CpFBA increased obviously in all wheat varieties in differentstress conditions. But, the expression patterns are inconsistent among several varieties or indifferent stresses of same variety.2. Afeter screening, there are18strains of positive strain T1generation transgenic plantdepartment; there are3strains of positive strain T2generation transgenic plant departmen.3. We get WCS120-CpFBA-eGFP plant expression vector.The study lay the foundationfor testing the function of CpFBA gene under the low temperature through genetictransformation methods.
