| The leaves,fruit,flower and roots in each part of the Pandanus tectorius Soland also know as wild pineapples which is the fruit of Pandanus utilis.Hainan has richer resources of Pandanus Linn.f,but the study for it is limited to the traditional medicinal efficacy and part of clinical medical research in our country.This article first studied the Pandanus tectorius Sloland of which fruits and nuts fat-soluble composition and content of total polyphenol technology,by using chromatographic column separating and appreciating preliminary discussed the chemical composition.This research first on doing preliminary experiment of the chemical composition for Pandanus tectorius Soland,combining with the resulrs,the key to study on the fat-soluble composition and total polyhenols of Pandanus tectorius Soland. Using the Folin-Ciocalteu colorimetry to explore the conditions of determination of total polyphenols in Pandanus tectorius Soland,and the total polyphenol content as an index, comparing the impregnation method,refiux extraction and ultrasonic extraction, determination of effective components of Pandanus tectorius Soland optimum extracting method and technological conditions, by using chromatographic column of silica for separating the ethyl acetate extract of Pandanus tectorius Soland1Quantitative analysis on effective constituents of Pandanus tectorius SolandAfter the quantitative analysis on effective constituents of system,Pandanus tectorius Soland contained alkaloids, steroids and triterpene, volatile oil, flavonoids, phenolic compounds and tannins (not found in the water extraction liquid, exists in ethanol extract solution). And quantitative analysis of the main active ingredients in Pandanus tectorius Soland indicate the moisture content is higher was57.33%, total polysaccharide content was27.7%, and total polyphenol content (1.597±0.07%) compared with total alkaloid(0.158±0.17%) and total metal content (1.235±0.13%) is higher, the content of total alkaloid is least.2The fat-soluble componentsi in furit of Pandanus tectorius Soland compared with in nutsBy soxhlet extraction method to extract the fat-soluble components of fruits and nuts of Pandanus tectorius Soland indcated:fruit and nuts of Pandanus tectorius Soland in the petroleum ether extract the content is difference bigger, the nuts of the content of45.35%, while only the fruit of4.91%. By GC-MS analysis showed, isolated from the petroleum ether extract of the fruit,11peaks were identified nine compounds, most of the fatty acids. In nuts isolated from the petroleum ether extract of five peaks, which identified three kinds of compounds, including with palmitic acid, linoleic acid and trans squalene, linoleic acid the relative content of65.58%, linoleic acid squalene relative content of18.62%.3Analysis the condition of determination of total polyphenol content in Pandanus tectorius Sol.We investigated the optimum reaction conditions of Folin-Ciocalteu colorimetry for the determination of Pandanus tectorius Soland used gallic acid as a reference. The results showed that when sodium carbonate solution concentration was10%, the Folin-Ciocalteu reagent was1.5mL, the Folin-Ciocalteu reagent to10%sodium carbonate solution(V/V)was3:4, reaction temperature was40℃, the reaction time was40min, the ccontent of total polyphenols had a good linear relationship. The methods for determining the average recovery was97.19%, relative standard deviation (RSD) was4.62%.4The different extraction method to extract the total polyphenols of Pandanus tectorius Solandand process optimizationBy the impregnation method,reflux extraction and ultrasonic extraction to extract the effective components in Pandanus tectorius Soland, to determination the content of total polyphenol as indicators, comparing extraction yield of three methods, the ultrasonic assisted extraction method can effectively extract the chemical compoent of Pandanus tectorius Soland.Combined with single factor test, the application of response surface analysis method for ultrasonic-assisted extraction method to extract the Pandanus tectorius Soland.The best extraction technology conditions:extraction temperature is53.46℃, time is26.82min, and solid-liquid ratio is46.40mL/g, total phenol content of the theoretical value of16.5232. To verify the feasibility of the response surface method, the choice for54℃temperature, extracting time for27min,47ml/g ratio of material liquid, extracting validation test for Pandanus tectorius Soland, parallel test three times to get the actual average total content was15.974mg/g, compared with the theoretical value of error is about3.32%.5The ethyl acetate ingredient separation of Pandanus tectorius SolandFrom Ethyl acetate extract of Pandanus tectorius Soland separated four monomer compounds,combined with the1H-NMR,13C-NMR spectroscopy methods, such as beta sitosterol, stigmasterol,vanillin and beta carotene. |
