Undaria pinnatifida, a kind of Perennial brown alsge, is the third largest economically cultivated seaweeds in China which has very high economic and pharmaceutical values. containing not only abundant proteins, vitamins and minerals, but also all kinds of unique physiological active components including alginic acid, mannitol, fucoidin, highly unsaturated fatty acid, fucoxanthin, organic iodine, sterol compound and dietary fibers etc.Most of the U.pinnatifida processings in China are in very initial stage, so the kelps are mainly exported to Japan as raw materials, U. pinnatifida in traditional industry was only used to produce low value products such as alginic acid, iodine and mannitol, but the exploitation and utilization of high value products such as fucoxanthin, fucoidin and fucosterol were seldom realized, which leads to the low efficiency of Undaria usage, causing serious resources waste and industrial development, restriction.In order to reduce the cost for Upinnatifida extraction process and improve its utilization efficiency as well as the product values, we studied the deep processing techniques and comprehensive utilization strategies for U.pinnatifida. Hoping to brings some sicentific and theoretical basis for U.pinnatifida industr in the future.First step, ultrasonic extracting fucoxanthin from U.pinnatifida by ethanol solvent. Affecting factors for fucoxanthin extraction technique were optimized by single factor and orthogonal analysis. Among the four affecting factors concerned, solvent concentration had the most significant effects on the extraction efficiency, and the other three had less effects which was extraction temperature, ulrasonic time and ratio of sample weight to solvent volume, according to the affecting capacities. the optimum extraction model is as follows:solvent concentration90%, extraction temperature40℃, extraction time30minutes, ratio of sample weight to solvent volume is10:1. By this technique, fucoxanthin extraction ratio was1.362mg/g (dry weight) by HPLC test.Second step, ultrasonic extracting mannito from utilized U.pinnatifida powder with ethanol. In order to gain optimization extract condition, we have optmized condition by single factor and ortogonal experiment. Finnal extraction model as follows:extraction temperature70℃, solvent concentration90%, ratio of sample weight to solvent volume10:1, extraction time20min. Productive rate of crude mnanito is around5.48%. We purified mannito through heavy crystallize, after purefy the pureness is97.6%.Third step, Fucoidan was extracted from utilized U.pinnatifida powder. In order to gain optimization extract condition, we have optimized condition by single factor and orthogonal experiment, eventual extract model as follows:extraction temeprature60℃, extraction time25minutes, ratio of sample weight to solvent volume50:1. Product rate of fucoidan was11.39%detected by Polysaccharide content.Fourth step, soldium alginate was extracted from utilized Undaria pinnatifida powder. In order to gain optimization extract condition, we have optimized condition by single factor and orthogonal experiment, eventual extract model as follows: sodium carbonate concentration3%, extraction temeprature50℃, and extraction time30minutes, atio of sample weight to solvent volume50:1. Production rate of soldium alginate finally is30.8%, viscosity is1.67Pa.s。According to the optimzed technique,100g raw U.pinnatifida (dry) can get136.2mg purified fucoxanthin,5.48g mannitol,11.34g fucoidin, as well as30.8g soldium alginate.
