Polysaccharide from Cordyceps gunnii(Berk.)Berk. was reported to possess several bioactivities, such as anti-oxdiant, anti-tumor, anti-aging activities and is able to increase immunity abilities. In this study, we performed chmical modifications (sulfation and carboxymethylation) of the polysaccharide to increase its bioactivities. The work provided new areas for the development and application of polysaccharides from Cordyceps gunnii (Berk.)Berk.In this study, the polysaccharide from Cordyceps gunnii (Berk)Berk. was obtained via water soaking and enthol precipitation from cordyceps sinensis, and named PS50and PS70. PS50and PS70were sulfated via using concentrated sulfuric acid method, obtained SPS50and SPS70, with the yields of62%and41%separately; and carboxymethylated via using monochloroacetic acid method,obtained CPS50and CPS70, with the yields of66%and54%separately. The purification of SPS50and CPS50were performed by using gel-filtration chromatography, with the purities of97%and86%.The sulfation percentage was0.33determined by gelatin turbid imetric analysis method, and carboxymethylated percentage was0.15determined by acid-wash method. The specific rotation were+28.33°,-36.75°,+12.5°for PS50, SPS50, CP50seperately. FT-IR, NMR, AFM, SEM, GC-MS were used to analysis the structure of SPS50and CPS50.We carried out the in vitro anti-oxdiant acitivity study on SPS50and CPS50. It was found that CPS50showed fairly good anti-oxdiant ability against DPPH. O2-and OH radicals, which is equal to PS50. SPS50showed equal anti-oxdiant ability against DPPH to PS50, but with lower anti-oxdiant ability against O2-and OH radicals compared to PS50. Anti-tumor experiment showed than the orignial PS50does not possess inhibition activity against K652tumor cells, whereas SPS50exhibited certain such activities. CPS also showed lower anti-tumor activities against K562tumor cells.
