Cordyceps sinensis (Berk) Sacc. is a traditionally rare Chinese nourishingherbs, has a remarkable effect in the aspect of enhancing the human immune systemand anticancer. The main extract is Cordyceps polysaccharide, can remove the bodyfree radicals, help the body delay aging, widely used in health care medicine andcosmetics fields demand. But natural Cordyceps yield is few, the price is expensive,difficult to meet the needs of industrial production.We study extraction method of liquid submerged fermentation culture ofCordyceps sinensis mycelia and Cordyceps polysaccharide, and focus on theultrasonic extraction of Cordyceps polysaccharide was improved, which lays thefoundation for the further development and utilization of Cordyceps polysaccharide.1. This paper uses orthogonal experiment to confirm the optimum cultureconditions of Cordyceps sinensis mycelia: cycle for7-8days, temperature is24-26degrees Celsius, pH is6.0-7.0, glucose is30g/L, peptone is10g/L, KH2PO4concentration is0.3g/L, Cordyceps mycelium and extracellular polysaccharide yieldwas the highest, mycelium yield was18.39g/L, the yield of exopolysaccharide was3.96g/L.2. Respectively, uses orthogonal experiments on the hot water bath extractionand ultrasonic extraction method, and selects the best extraction conditions, theoptimum extraction conditions of income under the orthogonal experiment, theultrasonic extraction of Polysaccharides from mycelium with hot water bathextraction polysaccharides compared, extraction time of the ultrasonic method wasshortened by96%than the hot water bath method, the yield of polysaccharidesincreased by2.9%. Analysis on antioxidant activity of polysaccharides obtained, thehot water bath extraction income Cordyceps much of hydroxyl radical scavenging raterespectively is68.24%,70.23%,69.66%, and the average removal rate was69.38%;the ultrasonic extraction method of Cordyceps Polysaccharide on hydroxyl radicalrate were59.48%,60.23%,58.78%, and the average clearance rate was59.50%. 3. On the basis ultrasonic extraction of Cordyceps Polysaccharide, added amountof KI to extract, to protect the cordyceps polysaccharide antioxidant activity ofobjective. In the cordyceps polysaccharide concentration under the condition of5mg/ml, the control group did not join the KI in the ultrasonic extraction method, theextraction of the Cordyceps Polysaccharide on hydroxyl radical rate is respectively58.61%,60.22%,59.38%, and the average removal rate reached59.40%, after addingKI in the extract, ultrasonic extraction the three group, repeat the income ofCordyceps Polysaccharide on hydroxyl radical rate is respectively64.81%,63.59%,65.36%, and the average removal rate of64.59%.The results show that adding KI, extraction of Cordyceps polysaccharide processas a protective agent can effectively improve the antioxidant activity of CordycepsPolysaccharide in ultrasonic method, the average rate of hydroxyl radical scavengingwas increased from59.50%to64.59%, as a result of the industrialization ofCordyceps polysaccharide extraction provides a certain reference.
