In this study, mitochondrial DNA COI and16S rRNA genes were used test on eight different natural populations of Hemifusus tuba (Gmelin) in coastal waters of China to reveals population genetic structure of H. tuba and discuss the possible mechanism. At the same time provide data for H. tuba fishery resources reasonably development and management in the future. On the other hand, fourteen polymorphic microsatellites were isolated firstly and characterized in Lutraria sieboldii, these markers will be useful for the study of population structure and genetic diversity of L. sieboldii. The main results are summarized as follows:Phylogenetic analysis based on NJ analysis for the COI and16S rRNA gene clearly divides the species into two lineages:northern lineage and southern lineage. The northern lineage composed of Lianyungang and Wenzhou population, and the southern lineage composed of Xiamen, Taishan, Zhanjiang, Beihai, Qinzhou, Fangchenggang population. Pairwise FST of COI and16S rRNA ranged from0to0.936and from-0.004to0.984, respectively. The FST values were significant both among northern and northern lineage and whitin populations.The results are basically the same with morphological variations analysis.Fourteen polymorphic microsatellites were firstly isolated and characterized. The number of alleles ranged from2to16per locus. The observed ranged from0.200to1.000, average0.609. The expected heterozygosities ranged from0.183to0.934, average0.607. These markers will be useful for the study of population structure and genetic diversity of L. sieboldii.
