Abstract/Summary:
The changing of the hormone, which effects on the growth of the follicle, gradually becomes a hot topic of the medical field and biological research in recent years. Many researches have already proved that reproductive performance has close relationship to the inner environment of the follicle. In this experiment, by doing the analysis of hormone in different levels in pig follicle, the localization and expression of the renin-aldosterone receptor in the testicular tissue and ovarian tissue of pigs and buffalos give us a primary impression on that how does the inner environment affect the growth and development of follicle. Their performance theories are basic on the further research of the ovarian inner environment.Experiment I:Collecting the follicular fluid by classifying the follicles of pigs’fresh ovary, and using their diameters as reference such as,2mm,3mm,4mm,5mm, and6mm. In order to measure the performance of hormonal underl5different level of the renin, angiotensin, aldosterone, progesterone, estradiol in different follicles with different diameters, we made the chemical fluorescence analysis. The result showed that the average diameter of the pig follicle had distinct positive correlation with the renin, aldosterone, estrogen and the progesterone in the follicular fluid. The tri-iodothyronine, dissociative T3, dissociative T4, thyroid binding globulin cortisol and the amount of the serum levels of ghrelin has no significant relationship with the diameters of the follicles. What’s more, the estrogen has distinct positive relationship with the progesterone in the follicular fluid of pigs. Besides, the Renin has the significant influence on the diameter of the hormones in the follicles.Experiment II:Colleting the fresh ovarian testicular tissue of buffalo and pigs and fixed them. Making the tissue sections after the two-step immunohistochemical staining and the results shows that renin receptor are evident expressed of ovarian follicle development at all levels in buffalo and pigs. Their positions are basically the same as well and they are mainly in oocyte and granulose cells. Buffalos’ and pigs’ testicles slices can be obviously observed that renin receptor expression. Testicles receptors located in the seminiferous tubules and spermatocyte at all levels. Thus infer pathways in the body’s endocrine, paracrine renin possibly through anticrime manner and gonadal tissue receptors, so that affecting the occurrence of gametes.Experiment III:Colleting of fresh ovarian and testicular tissues from pigs and buffalos. We made tissue sections after two-step immunohistochemical staining. The result shows:buffalos’and pigs’ovarian follicles slice of granulosa cells and significantly aldosterone receptor in the oocytes all had significantly expression, and they are showed in positive display. According to the results, we guess that:aldosterone receptors exist in the various stages of follicular development. There may also exist renin-angiotensin-aldosterone system in the ovarian and they play an important role in the development of granulosa cells and oocytes.
