In this study, a total of443canine brain suspected samples collected from Guangxi were detected by RT-PCR and six of them were rabies virus (RV) positive. The six rabies virus isolates were successfully isolated from the six positive samples by intracerebral inoculation of mouse. The isolates were named GXHCHJ, GXLZ04, GXRA03, GXRA06, GXLB19and GXNNSL, respectively. N and G genes of the six RV isolates were sequenced and phylogenetically analyzed by amplification of both genes. The results according to the their sequences showed that GXRA06, GXLB19and GXNNSL belonged to group! and GXHCHJ, GXLZ04and GXRA03belonged to Group Ⅱ.The rabies virus isolates GXLB19and GXNNSL, belonged to group Ⅰ, were conducted full length genome sequencing, as well as the isolates GXN119belonged to group Ⅲ, demonstrated that the length of complete genome of GXLB19, GXNNSL and GXN119were11921nt,11922nt and11924nt, respectively. While the full length of the isolate GXBH2011belonged to group Ⅱ was11924nt. Then comprehensive comparison and analysis were carried out with the isolates GXLB19and GXNNSL, GXBH2011, GXN119representing group Ⅰ, Ⅱ,Ⅲ respectively, and with that of other36rabies virus strains published in the NCBI from China and other countries at the complete genome level. As the results, The size of the individual structural gene, functional motifs and important functional sites and so on of the four rabies virus isolates in this study were consistent, only several sites differences. The mRNA expression affecting transcription initiation and termination signal sequences were extremely conservative, respectively AACA and G (A)7. In G-L non-coding region, the second conserved site TTP motif locating the470th nt of the downstream of the G protein stop codon was observed in the four isolates. In this study, measurement and comparative analysis complete genome sequences between different groups strains reflected the difference between different sources of rabies virus, enriched our background information, and provided a theoretical basis to develop effective prevention strategies or rabies vaccine for China.
