This study suspected rabies in Xinjiang Altay area three sheep brain tissue and forty-nine dog braintissue what were each Veterinary Bureau submission sample of Wensu, Qapqal, Jemnay, Haba River asexperimental material, using Direct immunofluorescence test, N gene of rabies virus specific purposeRT-PCR amplification and sequencing be to identify the virus. Using DNAStar-Lasergene.v7.1softwaresplicing sequence, BLAST software analyzed the N gene genetic evolutionary relationships. The resultsshowed as identify of the rabies virus for positive and obtained the virus complete gene sequences. Totallength is11923bp, the genome composition of the XJALT-2012-1virus genome sequence and otherpublished GenBank RV genome are basically identical. Genome-wide main structural protein codingsequence and size is3’-N (1353bp)-P (894bp)-M (609bp)-G (1575bp)-L (6387bp)-5’. There aredifferent length between structural protein gene of non-transcriptional gene spacer, N-P for2bp, M-P for5bp, P-Gfor5bp, G-L to423bp. Through XJALT-2012-1strain of five genetic evolutionary tree, five genesand their amino acid sequence variation frequency diagram and five hydrophobic amino acid sequencewere analyzed. Genetic relationship analysis show that the suspected rabies virus and the XJALT-2012-1virus genotype Ⅰ C group of Russian is the most recent, genetic relationship, belong to the sameevolutionary branches.Applied molecular bioinformatics software BioEdit to predicted protein amino acid sequence B cellantigen epitope regional of the XJALT-2012-1nuclear (N) protein and sugar (G) the advantage. Resultsshow that variation sites200、204-203of the XJALT-2012-1-G protein amino acids associated with thevirulence of rabies virus; The predominant epitopes of the B cell were probably in the N protein aminoacids regions of200、203、204and the G protein amino acids regions of130-138、191-201、505-520. Thisstudy provided a basis for further study toward molecular epidemiology of RV in China in the future.
