| The induction on embryoid of plant has many characteristics, such as, high reproductive rate,rapid growth rate and neat growth situation. However, it is time-consuming and difficult forthe embryogenic callus induction. That is to say, we just could obtain a small quantity ofgrapevine cultivars by this way. From the above, it is significant to increase the rate of theembryogenic callus induction and gain the regeneration plant. The anther, shoot tip, and stemtip of ruby seedless were used in the experiment. There was crucial effect of different trainingmethods and growth regulator combinations on induction of grape callus. The conclusionswere summarized as followed:1. Different training methods had different effects on induction of grape callus. On thesame condition, compared with liquid medium, solid medium had better results. When thegrape materials were training on the solid medium, the callus had high inductivity. Moreover,the average growth yield and proliferation multiple of solid medium were obviously higherthan liquid medium. When anthers were inducted on the solid medium, the average growthyield of callus was0.682, the average proliferation multiple was10.58, while on the liquidmedium, the average growth yield of callus was0.530, the average proliferation multiple was7.73. When shoot tips were inducted on the solid medium, the average growth yield of calluswas0.0122, while, the average growth yield of callus was0.0097on the liquid medium. Onthe solid medium, the average growth yield of callus of stem tips was0.0013, the averageproliferation multiple was0.85, while on the liquid medium, the average growth yield ofcallus was0.0010, the average proliferation multiple was0.59.2. The type of explants was important on the induction of callus and embryonic callus.The inducibility of different explants was disparate. On the induction of callus, the sort orderof inducibility was as followed: Anther>shoot tip>stem tip. And the highest inducibility ofanther was93.3%, the highest inducibility of shoot tip was76.7%, the highest inducibilityof stem tip was33.3%.On the induction of embryonic callus, anther and stem tip wereunable to induct embryonic callus, however, shoot tip could. The highest inducibility of shoottip was87.5%.3. Plant growth regulators influenced the induction of grape callus. When anther was used to induction callus, the optimal medium for inducing callus was NN69+1.0mg/L2,4-D+1.0mg/L6-BA; when the explant was shoot tip, the optimal medium was NN69+1.0mg/L6-BA; when the explant was stem tip, the optimal medium was NN69+2.0mg/L6-BA+0.1mg/L NAA.4. During the induction of embryogenic callus, NOA was crucial. The optimal mediumfor inducing embryogenic callus was X6+0.5mg/L NOA.5. On the aspect of germination rate and plantlet rate, the optimal medium for theembryoid to become integrated plant was GS+0.2mg/L IAA. |
