This test is mainly study on the total tanshinones and identified the mostappropriate method of extraction, isolation and purification. Systems extracttanshinones-β-Cyclodextrin for accessories, preparation of total tanshinones ofinclusion complex solution. Preliminary discussion of total tanshinones of inclusioncomplex solutions for several common pathogenic bacteria of dairy cow mastitis invitro anti-microbial activity. For developing treatment of mastitis of tanshinonespreparations provide some rationale.1. Visit heat reflux extraction and ultrasonic wave extraction of ethanol, respectively, as well as cold leaching extraction of three kinds of methods forextraction. With extraction rates as the main index, considering production costs andproduction time, thus extraction of determines the most suitable programmer.2. Comparison with macro porous resin adsorption purification and purification,water-alcohol extract of traditional heavy wash purification of Pharmacopoeiarequirements three purification methods, purification of total falconoid extractcontent and ultimately yield traits, to inspect and determine the best purificationscheme.3. Usaturated water saturated aqueous solution preparation of total tanshinonesextracted solution, respectively. Active ingredient content of inclusion complexsolutions for indicator preparation study on inclusion complex of temperature, time, materials and heating method. Method to determine the optimal preparation ofpackage of total ketene.4. Selection of Escherichia coli, Staphylococcus aureus, several kinds ofcommon pathogenic bacteria of dairy cow mastitis as test organisms. Flat filter1x108cfu/mL bacteria colony count method liquid coating plate, Cup and saucermethod for determination of antibacterial activity in vitro, agar dilution minimuminhibitory concentration determination. Preliminary study on inclusion complexsolution on in vitro antibacterial activity of several pathogens. Finalize a reflux of ethanol extraction of total tanshinones, suitable for the mostmanufacturers of veterinary drugs and equipment condition. Production costs are lowand short production cycles. Average extraction rate in90%(of tanshinoneⅡA as anindex).Water purification of extracts, purified extracts, content of tanshinoneⅡA in13.8%, cryptotanshinone in about5.2%. And total loss of one small before and afterpurification, is about23%. Purification operations simultaneously simple and lesstime consuming. Meet the requirements of industrial production.Use alternating hot and cold heating mode, to unsaturated aqueous solutionpreparation on the inclusion complex of extraction of total solution. Contents ofeffective components in inclusion complex solution for preparation of about3mg/mL.Than the other inclusion methods.Inclusion complex of Staphylococcus aurous Streptococcus, Bacillus subtitles andstopped milk suppression effect. It was found from the result that the totaltanshinones-β-cyclodextrin was active against all types of common pathogenicbacteria which were tested and exhibited a high sensitivity to taph.aureus, Bacillussubtilis and S.dysgalactiae equisimilis with the MIC of tanshinoneⅡA0.73μg/mL,cryptotanshinone2.51μg/mL;TanshinoneⅡA1.18μg/mL,Cryptotanshinone4.02μg/mLAt the same time, it had good solubility and had changed the poorlywater-soluble character of totaltanshinones, improved the weak absorption of totaltanshinonesd and raised the level of biologic utilization. So,this study can providesignificant basis for developing some biological agents and may instruct applicationof the antibiotics reasonably of the antibiosis drugs. It had brilliant prospects of utilityand was worth for further study.
