Sulfamonomethoxine(SMM)is a antimicrobial drug of strong effect, low toxicity,wide application and Broad-spectrum on fishery production.SMM have a good effect tobacterial vertical scale, vibriosis, gill disease and other diseases on aquaculture. Irratio-nal use of drugs not only cannot guarantee the efficacy of drugs, causing animal liver,kidney and other organ damage, but also pollute the environment to culture, and thedrug residues in aquatic products will threat the health of humanity. Therefore, it isnecessary to conduct the pharmacokinetic and residue analysis of SMM in tilapia. Inthis paper, by measuring the variation of concentration with time of SMM by HPLCmethod, we study the pharmacokinetics of SMM in tilapia. By studying the eliminateresidual rule of SMM comparatively at28±2℃、23±2℃respectively and differenttimes of administration, we expected to provide a scientific reference in the rational useof SMM on fishery production of tilapia. The main results of the study are as follows:1).The pharmacokinetic study of sulfamonomethoxine in tilapiaPharmacokinetics and elimination rule of sulfamonomethoxine (SMM) in tilapiawere researched by HPLC method after food feeding tilapia200mg/kg of SMM at28±2℃. Results showed that the Cmaxwere22.66,7.13,45.50and22.77μg/m L(μg/g),theTmaxwere7.52,7.02,1.00/8.00(Tmax1/Tmax2) and2.00/10.00h(Tmax1/Tmax2) in blood,muscle, liver and kidney of tilapia. Bimodal phenomenon of concentration-time curvewere found in liver and kidney,and the first peak was higher than the second,suggesting that there were Non-homogeneous absorption phenomenon in theGastrointestinal tract of tilapia. The Cmaxof SMM in the liver was highest in all thetissues while eliminated rapidly after the peak concentration, suggesting that first-passelimination occurs in the liver of tilapia. T1/2Keof SMM were5.21,4.84,14.12and6.8hin blood, muscle, liver and kidney which showed that SMM was short-acting sulfadrugs in tilapia and the elimination were faster in the muscle and blood than in the liverand kidney. 2).The comparison of residual rule of SMM in tilapia at different temperatures or admi-nistrationsWe feed tilapia by200mg/kg of SMM continuously for5days at28±2t℃and23±2℃. By measuring the variation of concentration with time of SMM by HPLCmethod, we study the metabolism of SMM in the blood and muscle of tilapia.Comparing to the results of pharmacokinetic experiments in the case of a single does tostudy the impact to the eliminate residual of SMM in tilapia at different modes ofadministration.The result showed that: the theory of times which the content of SMM in blood andmuscle of tilapia reduced to the residue limit(0.1mg/Kg) are160h and140hrespectively at23±2℃and they are135h,120h respectively at28±2℃; it showsthat the absorption and elimination of SMM in tilapia are accelerated with theincreasing of temperature. The theory of times which the content of SMM in blood andmuscle of tilapia reduced to the residue limits(0.1mg/Kg) after food feeding tilapia200mg/kg of SMM at28±2℃are130h and110h respectively and they are135h,120h respectively after feeding for5days at28±2℃; it shows that the residual timeunder the case of multiple dose will be extended than under a single dose and the reasonfor this phenomenon may be associated with the accumulation of SMM in animalsunder the case of multiple dose.Referencing the ordinance of the MRL of total sulfa drugs in foodstuffs of animaloriginal the notice No.235of china, the MRL of SMM is0.1mg/kg. According the testresults of residue in the muscle of edible tissues in tilapia, we suggest that therecommended withdrawal period is not less than6days after feeding SMM by200mg/kg.
