Bovine mastitis is an unsolved human challenge of the world. Although a lotof prevention and treatment of strategies were adopted, they didn’t change thissituation. HSP70i plays role in the activation of innate immunity, which isreceiving increasing attention lately. As a member of HSP70family, HSP70-2genepossibly contributes to animal susceptibility to udder inflammation. However, littleis known about HSP70-2polymorphism related to mastitis in Chinese Holstein.PCR-SSCP analysis was adopted to detect the polymorphism at thefragmentary of coding sequence region (1to1926)and5’/3’-untranslated regions(-170to0;1927to2127) of the HSP70-2gene in Chinese Holstein.25new SNPswere detected in this study:13SNPs in CDS region and12SNPs in UTR.2SNPsbring about amino acid changes in the mature HSP70-2protein: g.1770C→G(590Asp→Glu) in allele L and g.1877G→C (626Gly→Ala) in allele M. Mutationsin g.1877G→C made ghydrophilic amino acids (626Ala) turned into hydrophobicamino acids (626Gly) and influenced the protein secondary structure. Threepotential cow HSP70-2proteins were found in all the103individuals. However,the predicted three-dimensional structures of three potential proteins have nodifferences. Motif analysis showed that g.1877G→C (626Gly→Ala) led to losinga glycine in Glycine-rich region profile, and amino acid region corresponding withg.1743G→A, g.1746C→T, g.1761T→G that completely linked with g.1770C→G occur in Casein kinase II phosphorylation site. Statistical analysis of theassociation between genotypes and susceptibility of clinical mastitis indicated thatmorbidity rate of CC, JL, MO and MN is higher than other genotypes.9SNPscorresponding with this four genotype (g.-115G→A, g.-98T→C, g.117C→A,g.156A→C, g.1743G→A, g.1746C→T, g.1761T→G, g.1770C→G, g.1877G→C)were suggested to be associated with mastitis and can be used as molecular markersto breed the Chinese Holstein resistant to mastitis.
