Interleukin-8(interkeukin8, IL8), also known as neutrophil chemokine or neutrophil activation factor, is a member of the chemokine family.Test showed that there was CXCL8 with breast cancer in breast tissue pathological grade high expression of tumor metastasis, local recurrence negative correlation. And CXCL8 secretion in the serum of patients with mastitis is higher than the secretion of serum of healthy controls.CXCL8 research focuses on the human breast cancer, this experiment CXCL8 gene as the impact of milk production traits and mastitis candidate genes to explore with cows production performance and mastitis correlation.In this study, we use software DNAMAN, online software ExPASy and PSORT II Prediction to analysis the bioinformatics of CXCL8 gene in Chinese Holstein. And 446 blood samples were collected to extract genomic DNA, the genomic DNA of Holstein was used as template for amplify target sequence then sequencing the PCR product of target to detect the SNPs of candidate gene. Then genotype those SNPs with PCR-RFLP and Tetras-primer ARMS-PCR technique. Finally, performing its association with mastitis and milk production,which provided the genetic basis for the efficient selection of Chinese Holstein and the conservation and application of germplasm. The results were as follows:1. Bioinformatics Analysis of CXCL8geneHomology analysis of cows, sheep, pigs, dogs, rabbits, and Mustang shows that homology of cows, sheep and pigs gene are more than 90%, which shows the highly conservative among species. Structure and function of proteins predicted results show that CXCL8 protein is hydrophilic proteins in the endoplasmic reticulum presence of a small amount.2. Genetic variation detection of CXCL8geneA total of 4 exons and part of intronic were analyzed, eventually detect four SNPs site.They are located in the intron region.3. Population genetics analysisCXCL8 gene for population genetics analysis, C202 T, C264 T, A302 G loci are in moderate polymorphism, A2471 G is low polymorphism. C202 T and A302 G are inHard-Weinberg equilibrium(P>0.05), C264 T, A2471 G two mutations are not in Hard-Weinberg equilibrium(P <0.05).4. Analysis of the SNPs associated with milk production traits and mastitisUse SPSS 20.0 least squares analysis showed that A2471 G and cow milk fat was significantly associated(P <0.05), AG genotype has a lower fat percentage.Bioinformatics analysis, polymorphism detection and associated analysis with milk production traits and Mastitis of CXCL8 gene in Chinese Holstein provides a theoretical basis for the further study of the biological function of CXCL8 gene, and a theoretical foundation for understanding the mechanism of resistance in mastitis.
