The corn basal stem rot is a main disease of maize, In the spring maize area ofnortheast,north, northwest of China, Huang-Huai-Hai summer maize region andsouthwestern hills and mountains where maize growing,the occurrence iswidespread,and seriously affect the quality and yield of maize[1]. The maize stem baserot is a soil, seed-borne disease that infection by alone or mixed Pythium andFusarium. In order to grasp the culture and propagation technique of corn stem baserot bacteria in Anhui province, provide better technical service for ResistanceIdentification,this article studied the biological characteristics of corn basal stem rot inAnhui Province and had carried disease identification of validation on maize. Andalso provide a scientific basis for the disease resistance maize breeding,and therational distribution of disease-resistant varieties inAnhui province.The F.graminearum Schawbe is from two different places and the(P.aphanidermatum(Eds.)Fitzp is from three different places. This paper studied itsbiological characteristics and the virulence influence of different bacteria inoculationon the maize varieties; and carried on the resistant identification of19Introductionvarieties and74for the test for examination of varieties of Anhui Province, selectedsuitable resistant varieties for cultivation inAnhui Province.The main results are as follows:1、The optimization of corn basal stem rot medium formulationBy measuring the different culture methods have an effect on corn basal stem rot((P.aphanidermatum(Eds.)Fitzp) mycelial growth rate. The results showed that nosignificant difference in the mycelial growth rate of the strain in the two basalmedium of PDA and PCA; when the C source in the basal medium is sucrose,mycelial growth rate is highest, followed by starch, maltose and glucose; the N sourcein the medium is Ca (NO3)2, mycelial growth rate highest, followed by NH4NO3,NH4Cl; carbon content of sucrose in the basal medium is0.10%, the(P.aphanidermatum(Eds.)Fitzp mycelial growth is the best.2、Screening the solid medium of propagation of bacteriaSporulation instructions on solid medium were measured screening. The resultsshowed that on the corn solid medium,(P.aphanidermatum(Eds.)Fitzp sporulation ishigher than the other six kinds of solid medium; on the barley solid medium, F.graminearum Schawbe sporulation is higher than the other six solid medium.3、Comparison strain virulence of different regionsThe results showed that the virulence of F.graminearum Schawbe strainscollected in Fengyang Anhui is greater than the strains provided by the ChineseAcademy of Agricultural Sciences; the virulence of (P.aphanidermatum(Eds.)Fitzpstrains provided by the Chinese Academy of Agricultural Sciences, is greater than thestrains of Fengyang Anhui and Hebei Academy ofAgricultural Sciences.4、Germs solid culture was inoculated on the the corn stem base rot disease severityF.graminearum Schawbe and (P.aphanidermatum(Eds.)Fitzp solid cultureinoculum size is set to30g/strains,50g/strains,100g/strains,200g/strains,300g/strains five gradient. The results showed that the solid culture inoculated of twobacteria is200g/strains to300g/strains, the incidence and severity performance isthe most stable.5、Identification the resistant of19Introduction varieties and74for the test forexamination of varieties of Anhui Province, by two points repeat the test,The resultsshowed that:19varieties of introduction, the resistance of R (resistance) level andabove have5varieties, namely Jiaodan No.6, Suyu28, Yufeng3358, Dazhong858and Longping208, accounting for26.32%of the introduction and varieties; HS (highsensitivity) have4varieties, accounting for21.05%of the introduction and varieties.74for the test for examination of varieties, the resistance of R (anti) above have16varieties, respectively Xin’an No.15, Yuanyu906, Luyu9104, Aoyu3804, FengyuNo.6, SY1003, AN1010, Jinfeng No.8, Hedi328, Lvdan9118, HH9904, Woyu77,Wanjian11-3, Luyu9105, Jinzhai34and Aoyu3765, the test for examination ofvarieties2,162%; HS (high sensitivity) have11varieties, accounting for trialexamination of varieties of14.86%.6、Compared with countries Resistance Identification results, identification of theresistance of the varieties were similar in the two years of2007and2012, therespectively of resistance R (resistance) level and above varieties in the total speciesare22.58%,21.62%; the proportion of HS (high sensitivity) varieties are14.52%,14.86%; the proportion of S (sense) varieties are11.29%,13.51%.
