Watermelon (Citrullus lanatus) with a long cultivation history was a world-widehorticultural crop. It played an important role between in adjusting the agricultural structureand increasing the income of farmers. The core primers can be used for the analysis of geneticdiversity between various types watermelon, so it was applied to construct a moleculartechnology of DUS testing and DNA fingerprinting. this technology was used to analyzepopulation structure of788watermelon cultivars at the same time; A man-machine dialoguemode was constructed, it was a fingerprint database management system based on thecomputer system.1.23pairs of SSR primers and56morphological traits for DUS testing were used fordiversity analysis of24example varieties in watermelon DUS testing. There were73polymorphic loci, and the percentage of polymorphic bands was73.74%, the PIC valueranged from0.14to0.65, with an average of0.38.24example varieties can be divided into3class by23pairs of SSR primers on the genetic similarity coefficient of0.80. but it can bedivided into two groups by56morphological traits on the genetic similarity coefficient of0.66. The coefficient between two markers was0.186, it was not significance correlation.Nucleic acid fingerprint classification is more accurate than the traditional morphologicalclassification.2. two-dimensional (QR) code was used to built SSR fingerprints of the the watermelon24standard varieties,"Jingxin No.2" and "Jingxin No.1" were determined by DUSmolecular testing system, their similarity coefficient was0.89. Compared to themorphological identification,SSR markers had more differences locis.3. PowerMarker3.25software was used to analyze the genetic diversity of788watermelon cultivars,the result showed that they can be divided into East Asia ecotype,America ecotype, egusi and Citroides ecotype. STRUCTURE software were used forstructure population analysis of watermelon cultivars with every material blood composition.4.788Watermelon varieties was used to construct fingerprints database. The dialogueof human-computer can be achieved by computer software based database managementsystem.
