Arprinocid was a new anticoccidial drugs that belong to nucleosides. It has significant effect on Eimeria tenellum in poultry and no cross resistance with other anticoccidial drugs. Arprinocid can not be dissolved in the water, added it in diet may unevenness. However, that problem could be solved by arprinocid suspension which can diluted with water. Preparation of arprinocid suspension and its effects on Eimer tenella were studied in this paper.1. Preformulation Research of Arprinocid SuspensionA shake of flask-ultraviolet apectrophotometry method was established to determine octanol/water partition coefficient of arprinocid; an HPLC method was established to determine arprinocid API and arprinocid in suspension. The octanol/water partition coefficient results showed that1gP<1while pH<3and lgP>1while pH>4. The Silica Column(4.6mmx250mm,5μm)was adopted for HPLC, The detection wavelength was261nm, the mobile phase was isopropanol:methanol(75:25, v:v)and flow rate was0.8mL/min. A linear relationship existed in the calibration curve from10to80μg/mL,R2=0.9992, the mean recovery was99.7%. The average of inter-and intra-day precision (RSD) was1.1%. Stability test showed that the drug content is no significantly decline(≤5%), in line with the preparation requirement.2. Preparation And Technology of Arprinocid SuspensionThe Sedimentation rate and stabilize time were selected as the evaluation index to investigate the formulation and technological factors to arprinocid suspension stability. The prescription and preparation were optimized by orthogonal experimental design and Single factor. The results showed that the optimized formulation was composed of veegum(3%), CMC-Na(0.4%), sodium hyposulfite (0.25%), Methylparaben(0.2%), arprinocid(4%)and appropriate amount Tween-80. The preparation was conduct by ultrasonic400w, keeping10min in laboratory. Prescription and preparation selected was consistented with the requirement that instituted by Pharmacopeia and displayed a better stability. 3. Physicochemical Property And Stability Of Arprinocid SuspensionAn accelerated tability testing and long-term tability testing, physicochemical property, particle size, pH, viscosity and contents of arprinocid suspension were investigated.The results showed that the average particle size was0.77μm,the pH was5.43, the viscosity was increasing while the shear rate was decreasing, indicating that the suspension was belong to non-Newtonian fluid. The stability tests showed that freezing thawing tests had a significant influence on stability of this suspension. However, the illumination had no effect on it. The appearance,content, stabilize time and physicochemical property of suspensions maintained in a good condition for the three month in (30±2)℃conditions or at room temperature, indicating that the suspension consistented with the requirement that instituted by Pharmacopeia.4. Efficacy Of Arprinocid Suspension Against Eimeria Tenella In Chicken.Efficacy of arprinocid suspension againse eimeria tenella was tested in chicken.Artificially infected with Eimeria tenella of5×104orally per chick. All the chicken were diveded into six groups, In three group, the levels of arprinocd were20mg/L,40mg/L,60mg/L Toltrazuril-treatment group (25mg/L),the healthy control group and unmedicated group, The results showed that the ACI of the20mg/L,40mg/L,60mg/L of arprinocid suspension and Toltrazuril group were159.1,178.7,167.3,193.3, respectively, indicating medication with40mg/L could control eimeria tenella well.
