Citrus paradise is known as the grapefruit or Fan citrus, and is belongs toRutaceae Citrus of evergreen paints. Grapefruit is an economically important fruit with softjuice, unique flavor and favorable taste. It can be consumed with fresh pulp or process. Thepeel contains a variety of mineral composition, and seeds can be extracted of essential oil.In recent years, the fruit is a treasure and widespread concern of consumers or farmers athome and abroad. However, the optimal root stock matching is a major bottleneck in thegrapefruit research. Enzyme as the direct product of the plant genes, which has been usedto as a predictor of graft affinity in many plants grafted, but is blank in the study ofgrapefruit.Nine varieties of grapefruit introduction from U.S.A were grafted with four rootstocks in the research. By measuring POD, SOD, CAT, PPO, and PAL activity ofcombination with different scion healing process in grafted, to learn about the differenceenzyme species and activity role between roorstock. Establish the screening systembetween physiological activity and affinity. The preliminary study was provided foroptimal combination of scion in protein levels. The thus the most suitable rootstock forperennial fruit trees early matching scientific and theoretical basis. The main results aresummarized below.1. The POD, SOD, CAT, PPO, and PAL activity are related with graft affinity ofgrapefruit, it can be used as early pre-selected indicators of grapefruit grafted affinity.Little effect between various enzyme activities furthermore different enzymes haveindependent influence in graft healing process.2. The absolute value between rootstock and scion enzyme activities was related tohealing before grapefruit grafted healing, and the correlation in different seasons of thesame enzyme were different. The closer of absolute value between stock and scion’senzyme activity of POD, SOD, CAT, and PPO before grafted healing in spring ofgrapefruit, the better of healing, but activity of PAL was no significant effect to healing. Inautumn, the closer of absolute value of enzyme between rootstock and scion activity ofPOD, SOD, CAT, and PAL before grafted healing of grapefruit, the better of healing, butactivity of SOD was no significant effect to healing.3. The grapefruit grafted healing parts enzyme activities in different seasons are showthat the activity of POD, CAT, and PAL in healing parts was higher can promote to grafthealing and improve the survival rate. In contrast, the activity of SOD and PPO in healingsite as low as possible. 4. The grafted survival rate related to enzyme activity had difference between thedifferent group of stock and scion. The grafted survival rate of C. mansailuo and C. grandisaurantium is significant correlation to the activity of POD, PAL. The grafted survival rateof C. mansailuo and C. aurantium is significant correlation to the activity of SOD, PAL.The grafted survival rate related to enzyme activity is not significant when the healing ofthe grafting of Poncirustrifoliata and Citrus paradise. All combinations of the CAT, PPOacticity and graft survival rate of the correlation was not signigicant.5. The comparison in different combinations affinity: In spring grafted, the affinitiesare shown that C. mansailuo has the best graft affinity in the nine varieties of grapefruit, C.aurantium and C. grandis aurantium are the second, the Poncirustrifoliata affinity is worst.The affinity of1#,2#scion and Poncirustrifoliata is superior to the other three rootstocks.The affinity of3#scion with C. mansailuo and Poncirustrifoliata is superior to the othertwo rootstocks. The strongest graft affinity is4#scion and Poncirustrifoliata, the next is C.aurantium or C. grandis aurantium, but the C. mansailuo is worst. The strongest graftaffinity is5#scion and C. aurantium, the next is C. aurantium or C. grandis aurantium, butthe C. mansailuo is worst. The6#scion with C. mansailuo and C. grandis aurantium hasthe better graft affinity than others. The7#scion with C. mansailuo has the better graftaffinity than others. The8#scion with C. aurantium has the better graft affinity than others.The9#scion with C. mansailuo, C. grandis aurantium, and Poncirustrifoliata have thebetter graft affinity than the C. aurantium.In autume grafted, the tendencies of the grafter affinity are same with the springaffinity. The affinity of1#,4#scion and C. mansailuo is superior to the other threerootstocks. The3#scion grafted affinity with the Poncirustrifoliata, C. aurantium and C.grandis aurantium is superior to the C. mansailuo.5#scions with C. grandis aurantiumgrafted affinity better than the other three rootstocks. The affinity of6#,7#,8#sciongrafted with C. aurantium, C. mansailuo and C. grandis aurantium respectively have thebest effect. The9#scion with C. mansailuo, C. grandis aurantium, and Poncirustrifoliatahave the better graft affinity than the C. aurantium.All analysis indicated that the value of POD, SOD, CAT PPO and PAL activities can beused as early pre-selected indicators of grapefruit grafted affinity. The affinity of differentscions with four rootstocks has the best effect in C. mansailuo, the next is C. aurantium orC. grandis aurantium, the Poncirustrifoliata is the worst. As the condition of Puwenforestry farm, the best effect of affinity combination be confirmed as:6#scions with C.mansailuo, C. aurantium;7#scions with C. mansailuo;8#scions with C. grandis aurantium;9#scion with C. mansailuo, C. grandis aurantium, and Poncirustrifoliata havethe better graft affinity.
